Seroprevalence And Genotyping Of Toxoplasma Gondii Infection In 729 Reproductive-age Women In Guiyang

Objective: To investigate the seroprevalence and genotyping of Toxoplasma Gondii(T.gondii)infection in 729 women of reproductive age in Guizhou province.It is to provide the prevention and treatment of toxoplasmosis infection for women of reproductive age in guizhou province. Methods: The enzyme-linked immunosorbent assay(ELISA) was applied to test the specific antibodies(IgG, IgM)and circulating antigens(CAg)of T.gondii in 729 women of reproductive age.Moreover, the 529 bp specific repeat DNA of T.gondii was investigated by Polymerase Chain Reaction(PCR). The genotyping of T. gondii was determined by multiplex multilocus nested polymerase chain reaction–restriction fragment length polymorphism(Mn-PCR-RFLP). Results: Among 479 pregnant women, the positive rate of CAg IgG, IgM was 0.63%,7.1%,0.84%.Separately, the positive rate of CAg IgG and IgM in normal pregnancy group was 0.56% 4.47% 0.56% and0.83%,14.87%,1.65% in abnormal pregnancy group. Meanwhile,The positive rate of IgG was 4 % in 250 healthy people. The IgM and CAg detections were negative in all healthy people samples. Analysis of the serological results of three groups, the difference between abnormal pregnancy group and normal pregnancy group was statistically significant(χ2=16.003, P<0.05). The statistical difference of abnormal pregnancy group and healthy people was significant(χ2=18.992, P<0.05). There was no difference in normal pregnancy women and healthy people(χ2=0.790, P >0.05).The 529 bp specific repeat DNA of T.gondii was found in three samples.One belongs to normal pregnancy group,two belong to abnormal pregnancy group.The genotyping was ToxoDB#9(Chinese 1).Conclusion: The main seroprevalence of Toxoplasma Gondii(T.gondii)infection in women of reproductive age may occur in those chronically infected.. The recent infection or acute infection are rarely appeared.It suggested that T.gondii infection is closely related to the abnormalpregnancy. Chinese 1 Toxoplasma may be a main lineage among the women of reproductive age in GuiZhou province. During the Clinical prenatal diagnosis period people should improve the recognition and diagnostic level.
