With the increasing types of domestic traditional Chinese medicine and Western medicine, combination of Chinese and Western medicine is widely used. It can both to improve clinical efficacy and reduce the adverse reaction which has been widespread recognition by the medical profession. This study through the animal experiments to explore the inhibitory effects of traditional Chinese medicine -extract of Pteris semipinnata with 5-FU on the growth of tumor, to reveal the interaction between traditional Chinese medicine and chemotherapy drugs and provide a theoretical basis to the use of combination with Chinese medicine and chemotherapy drugs on clinical tumor therapy.Methods1 Prepare Pteris semipinnata L. water extract and 65% ethanol elution and 95% ethanol elution, and determine the content of 5F in the extracts by HPLC.2 Establish model on Kunming mice by inoculating subcutaneously S180 mouse sarcoma cell line. Mice were randomly divided into negative control group〠positive control group of fluorouracil (15mg/kg)ã€Pteris semipinnata L.65% ethanol extract(300mg/kg)ã€three dose groups of Pteris semipinnata L.95% ethanol extract(150mg/kgã€300mg/kgã€600mg/kg)ã€Pteris semipinnata L.95% ethanol extract(300mg/kg)+fluorouracil (15mg/kg) groupã€Pteris semipinnata L. aqueous extract group (0.01mg/kg)ã€Pteris semipinnata L. aqueous extract (0. Olmg/kg)+fluorouracil (15mg/kg) group,9 groups in total,10 mice in each group. Administrate for 10 days continuously and observe the general conditions of the mice.24h after the last drug administration, weigh and kill the mice by taking off the vertebral. Cut and weigh the tumor〠spleen〠thymus and calculate the tumor growth inhibition rate.3 Establish model on Kunming mice by inoculating S180 mouse ascites cell line. Mice were randomly divided into negative control groupã€positive control group of high fluorouracil (15mg/kg)ã€positive control group of low fluorouracil (5mg/kg)〠Pteris semipinnata L.65% ethanol extract (300mg/kg)〠Pteris semipinnata L.65% ethanol extract (300mg/kg)+fluorouracil (5mg/kg)〠Pteris semipinnata L.95% ethanol extract (150mg/kg)〠Pteris semipinnata L. 95% ethanol extract(150mg/kg)+fluorouracil (5mg/kg)〠Pteris semipinnata L. aqueous extract group (0. 01mg/kg)ã€Pteris semipinnata L. aqueous extract (0.01mg/kg)+ fluorouracil (5mg/kg),9 groups in total,10 mice in each group. Administrate for 10 days continuously and observe the general conditions of the mice to twentieth day. Compare the survival time and calculate the life span.4 Establish model on Kunming mice by inoculating Ehrlich mouse ascites cell line. Mice were randomly divided into negative control groupã€positive control group of cyclophosphamide (20mg/kg)ã€Artesunate intraperitoneal injectin group(90mg/kg)〠Artesunate intramuscular injectin group(90mg/kg)〠cyclophosphamide + Artesunate,10 mice in each group. Administrate for 15 days continuously and observe the general conditions of the mice to twentieth day. Compare the survival time and calculate the life span.Results1 The content of 5F in Pteris semipinnata L. aqueous extract Pteris semipinnata L.65% ethanol extract and Pteris semipinnata L.95% ethanol extract was 0.21%ã€9.36%ã€2.95%。2 The extracts of Pteris semipinnata L. has significantly inhibition on Kunming mice bearing S180 sarcoma tumor.which has reached more than 30%. The inhibition rate of the extracts of Pteris semipinnata L. combining with fluorouracil was higher than monotherapy in a certain extent.Moreover the weight of spleen gland and thymus were heavier than the positive control group. Thus we can make the conclusion the extracts of Pteris semipinnata L. presumably protect the spleen glandã€thymus immune organs so as to achieve the purpose of anti tumor.3 Pteris semipinnata L. aqueous extract and Pteris semipinnata L.95% ethanol extract have a certain life prolonging on S180 ascites tumor bearing mice. And the life extension rate of Pteris semipinnata L.95% ethanol extract combination with low dose of fluorouracil is almost equal to high dose of fluorouracil.4 Artesunate on EAC ascites tumor bearing mice have a certain life-prolonging effect and the life extension of Artesunate combination with cyclophosphamide has increased to some extent than the two drugs use alone.ConclusionThe results showed that Pteris semipinnata L. has a antitumor activity on a certain extent. Moreover, the combination of the of Pteris semipinnata L. and fluorouracil has a synergism effect.
