Objective:To prepare lactic-co-glycolic acid (PLGA)/calcium phosphate cement (CPC) microcapsules and rhizoma drynaria poly(lactic-co-glycolic acid) (DR-PLGA)/Calcium phosphate cement (CPC) composite scaffold respectively;UV assay DR-PLGA/CPC scaffold drug loading, encapsulation efficiency, observe the effect of PLGA/CPC scaffold and DR-PLGA/CPC composite on treating rabbit femoral bone defect.Methods:1、Producing PLGA/CPC microcapsule and DR-PLGA/CPC composite by ultrasonic emulsification, refer to the relevant literature to determine the optimum process parameters.2、Eight Newzealand rabbits were randomly divided into two groups.Preparation of rabbit femoral defect model, the experimental group were implanted DR-PLGA/CPC composite while the control group PLGA/CPC Scaffold were implanted.After4,8 weeks,we took out the materials,observed with X-ray, gross anatomy, histology observation and electron microscope scanning to evaluate the osteogenetic activity, the effect of accelerating the healing of bone defect.Results:After surgery all animals achieved good recovery and there’s no sign of wound infection and death. The average entrapment rate of scaffold measured was 9.13%, The average drug loading rate of scaffold measured was 95.8 mg, At 4W and 8W after implantation,the effect of promoting fracture healing and osteogenic activity of the experiment group were greater than those in the control group.Conclusion:DR-PLGA/CPC composite scaffold and PLGA/CPC scaffold in rabbit bone defect healing process, can provide scaffold for osteoblasts, thus to promote bone defect healing; DR-PLGA/CPC composite promote the healing of rabbit femoral defect better than PLGA/CPC scaffold group, expounded Drynaria can effectively cure clinical treatment of bone defects.
