Objective:To investigate the effect of Jianpi Huayu Decoction (JHD) on the cell apoptosis and cell cycle in Human Colorectal Cancer Cells (SW480) and its mechanism, and further elucidate the possible mechanism of JHD inhibiting colorectal cancer growth and promoting apoptosis from the point of view of cell cycle and apoptosis, providing a new practicaltherapeutic strategy aga-inst colorectal cancer.Methods:1. MTT was used to determine the proliferation of SW480 cells with different doses of JHD at different times.2. Flow cytometry assay was used to determine the effects of JHD on cell cycle and apoptosis in SW480 cells.3. RT-qPCR assay was used to observe the expressions of genes (cyclinDl, cyclinE1、 CDK2, CDK4, CDK6, p27, Bax, Bcl-2) after JHD acting on SW480 cells.4. Western Blot assay was used to observe the expressions of proteins (cyclinDl, cyclinD2, cyclinD3, cyclinEl, CDK2, CDK4, CDK6, p27, Bax, Bcl-2, caspase-3, cleaved caspase-3, PARP, cleaved PARP) after JHD acting on SW480 cells.Results:1. JHD inhibited the proliferation of SW480 cells in a concentration and time dependent manner.2. After JHD acting on SW480 cells, it was found to have a strong apoptosis-inducing effect and induce G0/G1-phase cell cycle arrest.3. After JHD acting on SW480 cells, the gene expressions of cyclinDl, cyclinEl, CDK2, CDK4, CDK6, Bcl-2, and p27, Bax increased.4. After JHD acting on SW480 cells, the protein expressions of cyclinDl, cyclinD2, cyclinD3, cyclinE1, CDK2, CDK4, CDK6, caspase-3, PARP, Bcl-2 decreased, and p27, cleaved caspase-3, cleaved PARP, Bax increased.Conclusions:JHD may inhibit proliferation in human colorectal carcinoma cells (SW480) by inducing G0/G1-phase cell cycle arrest and apoptosis by downregulating the gene or protein expressions of cyclinD1, cyclinD2, cyclinD3, cyclinE1, CDK2, CDK4, CDK6, caspase-3, PARP, Bcl-2, and upregulating the gene or protein expressions of p27,cleaved caspase-3, cleaved PARP, Bax.
