Zanthoxylum dissitum was the raw material of many Chinese traditional medicine,due to its function of internal heat,fever and rheumatic pains. The existed analyzation about Zanthoxylum dissitum’s constituent and physiological features were fewer than the expected.In consequence, Zanthoxylum dissitum couldn’t be properly used.Poor growing environment,slow groth and low reproductive rates,result in the shortage of it and people’s over-exploittation make it worse.This paper analysed the relevant process conditions in preparation of Zanthoxylum dissitum and researched its physiological features. The main results were as follows:①Single factor experiment was used to find out four factors and three levels for orthogonal experiment to determine the total alkaloid extraction, the processes were as follows:Zanthoxylum dissitum powder was soaked in 20 times volume of 80% ethanol for 24h,then ultrasonic for 1.5h at 50℃.After filtration,the extracting process was repeated.The total amount of alkaloid extraction was 11.82mg/g. NKA-9 resin was selected to separate and purificate the total alkaloids,the specific conditions were: Zanthoxylum dissitum and resin weight ration was1:1(g/g),the sample was adjusted to pH 7.0 with flow rate 2BV/h.6BV and 80% ethanol were eluted with the speed of 2BV/h,and the purity of alkaloid was 32.9%.② The factors affect the total flavonoids were settled after single factor experiment and orthogonal experiment.They were:15times volume of 70% ethanol,24h soakage,50 ℃ ultrasonic lasted for 1.5h.The extracting steps were done twice.And the maximum extract was 4.78mg/g. AB-8 resin was selected to separate and purificate the total flavonoids.The isolation and purification conditions were:the consistence reagant was 1.5mg/mL with pH value of 6.0,and the flow rate was 2.0mL/min.After the drastic and dynamic adsorptive process in elution with 70% ethanol 140mL at the rate of 2.0mL/min.The purity of total flavonoids was 69.57%.③The optimum conditions for extracting morkit were:Zanthoxylum dissitum powder was soaked in 15 times volume of 50% ethanol for 24h,and extracted twice by ultrasonic at 60℃ for 40min.The morkit extraction could be 1.59mg/g.It turned out that S-8 resin was the most ideal material for separation and purification when morkit extraction’s pH value was 9.0 and concentration was 0.8mg/mL in dynamic adsorption,after 2BV/h dynamic adsorption,80% ethanol was used in elution at the speed of 3BV/h.The purity of morkit was 45.5%.④ kirby-bauer disc agar diffusion test and MIC proved that total alkaloid strongly inhibited staphylococcus aureus and colibacillus when MIC was 0.8mg/mL and 0.4 mg/mL separately.Compared to colibacillus, staphylococcus aureus was suppressed easily by total flavonoids when MIC was 1.6 mg/mL and 0.4 mg/mL separately.While morkit should be used in higher accumulation to suppress them.⑤ The study of DPPH and elimination ratio of hydroxyl radical,the antioxidant ability of them were as follows:total alkaloid> total flavonoids> morkit,which could offer some help for developing nature antioxidant.
