Objective To investigate the influence of sensitivity to radiotherapy, DDP chemosensitivity and tumor formation in nude mice for TCF21 (transcription factor 21) gene on human lung adenocarcinoma cancer A549 cells. Methods Using lentivirus-mediated overexpression of technology in human lung adenocarcinoma A549 cells to highly expressed TCF21 genes, adopting Fluorescence quantitative PCR (RT-PCR) and Western Blot technology verification the expression of target gene TCF21; MTT assay to detect the influence of sensitivity to cisplatin (cis-Dichlorodiamineplatinum, DDP) chemosensitivity for lung adenocarcinoma A549 cells which highly expressed of TCF21 gene; colony assay to investigate the influence of sensitivity to radiosensitivity of lung adenocarcinoma A549 cells which highly expressed of TCF21 gene; after lentivirus infection in each group (experimental group, negative control group and blank control group) were seeded in BALB/c nude mice subcutaneously the left armpit, nude mice to observe the situation and growth curve, Calculate the volume inhibition rate. Results TCF21 were successfully highly expression in A549 cells. at 72 h, with the increasing concentration of DDP (0,0.625,1.25,2.5,5,10 mg/L), each group corresponding inhibition rates increased, and the inhibition rate of the high expression group was significantly higher in empty vector group and untransfected group (P <0.05), no significant difference between the two then; overexpression TCF21 group with drug concentration and time and increase the rate of high expression inhibition corresponding increase (P<0.05); after receiving X radiation, non-transfected group, untransfected plus radiotherapy group, vector group, vector plus radiotherapy group, high expression and high expression+ radiotherapy colony formation rates were:95.17%±2.85%,88.20%±2.03%, 93.80%±4.17%,85.60%±2.42%,71.67%±3.21%,56.00%±2.65%; In nude mice experiment, the high expression group performance slow tumor growth compared with empty vector group and control group, tumor final volume was (620.08±263.07) vs (1406.77±351.14) vs (620.08±263.07) mm3 (P<0.05).Conclusion TCF21 gene expression can significantly enhance the A549 lung cancer cells to radiotherapy and chemotherapy DDP sensitivity; can effectively inhibit the growth of human lung adenocarcinoma A549 cells in nude mice transplanted tumors, indicating TCF21 play an important role in the development of human lung adenocarcinoma, and there may be a potential new target for therapy targeting human lung adenocarcinoma.
