Objective :To establish subcutaneously transplanted tumor of glioma models in nude mouse with green fluorescent protein(GFP), which is suitable for HBO therapy combining with radiotherapy in specific pathogen-free(SPF) environment, and investigate the biological characteristics.Methods :Totally 20 Foxn1 nu.B6-CAG-EGFP/SU mice were randomly divided into blank control group(C), radiotherapy experimental group(R), HBO combining with radiotherapy experimental group(H+R), respectively 8 mice in blank control group,6 mice in radiotherapy experimental group and HBO combining with radiotherapy experimental group.The radiation treated animals(12 mice)received a therapy of 10 Gy for three times with a total dose of 30 Gy, respectively on 7d, 14 d, 21 d after the injection. In HBO combined with radiotherapy group, The HBO treated animals(6 mice)received a therapy of 2.0 ATA with 100% O2 and maintained for 1h, started on 7d after tumor inoculation, lasted for 3 weeks, radiotherapy was implemented within 0.5h after HBO therapy with the same radiation dose and frequency as radiotherapy group.Result:The tumor masses obtained at the end of the experiments in the control group, radiotherapy group and hyperbaric oxygen combined with radiotherapy were 3115.625 ± 778.726(mg),1833.1667 ± 99.804(mg),1794.667 ± 152.913(mg) respectively. Tumor growth inhibition in combination group was higher than those in radiotherapy group and control group(P <0.01).Conclusion:Subcutaneously transplanted tumor of glioma in GFP nude mice was suitable for hyperbaric oxygen therapy combining with radiotherapy, which was good for evaluation of therapeutic efficiency.
