Objective:To observe the influence on the expression of renal tissues of BMP-7, MMP-2 and TIMP-2 in rats of chronic renal failure with 5/6 nephrectomy surgery by TaoHeChengQi Decoction and discuss the mechanism about the treatment of TaoHeChengQi Decoction for chronic renal failure, providing ideas and experimental basis for clinical treatment of chronic renal failure.Methods:60 male SD rats were randomly divided into blank group, sham operation group, model group, control group and treatment group, each with 12 rats. The blank group was without surgery, but the sham operation group was only with doing kidneys film peeling surgery. And the remaining three groups were with doing 5/6 nephrectomy surgery, establishing the CRF rats model.4 weeks after surgery, the treatment group and the control group were respectively given with TaoHeChengQi Decoction and NiaoDuQing particles. And the remaining three groups were respectively given with the same amount of physiological saline. After 8 weeks, blood serum were collected by abdominal aortic method, in order to detect the RBC. HB, serum Scr, BUN and serum 2-MG in rats. And urine were collected for detecting 24h urinary protein, urinary β2-MG, urinary NAG and urinary RBP. Taking kidney tissues of each group in rats is to respectively detect the expression of kidney tissue protein of BMP-7, MMP-2 and TIMP-2 and kidney tissue mRNA of BMP-7 and MMP-2 and to observe pathological changes.Results:(1)The RBC count and HB content in the model group were evidently reduced, while the serum Scr, BUN. serum P2-MG,24-hour urinary protein excretion, urinary p2-MG. urinary NAG and urinary RBP were significantly increased(.P<0.05). And the expression of the kidney tissue protein and mRNA of BMP-7 and MMP-2 were significantly decreased, while the expression of the kidney tissue protein of TIMP-2 was increased obviously(P<0.05). We can see the obvious proliferation of renal interstitial fibrosis under the light microscopy.(2)The RBC count and HB content were evidently increased in the treatment group and control group, but the serum Scr, BUN, serum β2-MG,24-hour urinary protein excretion, urinary β2-MG, urinary NAG and urinary RBP were clearly reduced(P<0.05). And the expression of the kidney tissue protein and mRNA of BMP-7 and MMP-2 were apparently increased, while the expression of the kidney tissue protein of TIMP-2 was obviously decreased(P<0.05). The two groups in the light microscope can be found in renal interstitial fibrosis lighter than the model group. In addition to routine blood, the treatment group was apparently better than the control group(P<0.05).Conclusion:(1)The TaoHeChengQi Decoction can relieve renal anemia, promote the excretion of metabolites, improve the function of glomerular and tubular and reduce the lesions of renal tissue, thereby delaying the time of chronic renal failure.(2)The treatment mechanism of chronic renal failure with TaoHeChengQi Decoction is by raising the expression of the kidney tissue protein and mRNA of BMP-7 and MMP-2, and through reducing the expression of the kidney tissue protein of TIMP-2, in order to reduce excessive ECM deposition, thus delaying renal interstitial fibrosis.(3)In addition to routine blood, the efficacy of TaoHeChengQi Decoction in treating chronic renal failure is better than NiaoDuQing particles.
