| Purpose:Through the animal experiment research Taohechengqi decoction on chronic renal failure(CRF)rats in the process of renal fibrosis p-GSK-3 β regulation function,to explore the mechanism of action of Taohechengqi decoction delay CRF,so as to provide theoretical basis for clinical treatment of CRF.Methods:1.Establish the CRF rats model:100 grade clean male Wistar rats,7 weeks,weight 180-200-g,adaptive breeding 1 weeks after buying back,were randomly divided into normal group,control group,model group,treatment group of 25 only.The model group and the treatment group adopted the international 5/6 nephrectomy animal model and performed 5/6 nephrectomy.In the sham operation group,only bilateral renal capsule was dissected;Normal group can’t operate.After surgery,penicillin was injected into the abdomen for 3 days.The serum Scr was collected from all the rats in three weeks after operation to observe whether the model was successful.nephrectomy.In the sham operation group,only bilateral renal capsule was dissected;Normal group can’t operate.After surgery,penicillin was injected into the abdomen for 3 days.2.After the success of the model.4 weeks after the operation,the medicine was given at 9 am every day.The treatment group was treated with Taohechengqi decoction.and the normal group,the sham operation group and the model group were given the same amount of physiological saline.The dosage of the rats was 6 times the weight of adults per kilogram,and the dosage was 8 weeks.To observe and record the general condition of the rats during the administration of the drug,once a week,to weigh the weight and adjust the dosage according to the latest weight,to record the death of the rats and the reasons for the analysis.3.The blood of the abdominal aorta was collected on the 57th day after administration,and the renal function(Scr,Bun)and blood routine(Hb,RBC)were detected.After the blood collection was completed,the residual renal tissue of rats was removed and divided into two parts.Some kidney tissues were placed in the frozen storage tube,and Western blot was used to detect the protein content of Wnt4,gsk-3 β,p-gsk-3 β and β-catenin.Another part of the renal tissue was preserved in a polyformaldehyde solution,which was used to observe the pathological morphology of the kidney after HE staining.Results:1,Compared with the blank group and control group,the Scr and Bun content of model group increased significantly(P<0.05),content of Hb,RBC count(P<0.05),Wnt4,p-GSK-3β,β-catenin significantly increased(P<0.05),no obvious difference in GSK-3β protein(P>0.05),then under the light microscope,glomerular contraction,basement membrane thickening,tubular dilation,structural disorder,renal interstitial fibrosis,inflammatory cell infiltration.2.Compared with model group,the Scr and Bun content,of treatment group rats were decreased(P<0.05),the content of Hb,RBC count(P<0.05),Wnt4,P-GSK-3β,β-catenin protein content decreased significantly(P<0.05),no obvious difference in GSK-3β protein(P>0.05),and then observe the results compared with model group the glomerular sclerosis,renal interstitial fibrosis hyperplasia was significantly lighter.Conclusion:1.The Taohechengqi Decoction reduce the Scr and BUN of CRF rats and to some extent,correct the anemia status of CRF rats,effectively improve renal function,and delay the process of renal fibrosis.2.The mechanism of Taohechengqi decoction in the treatment of CRF may be related to the regulation of Wnt4/β-catenin signaling pathway and the reduction of Wnt4,p-GSK-3β andβ-catenin protein expression. |
