Objection:The purpose of this study is to analysis of etiology and pathogenesis of ankylosing spondylitis by observing the Chinese herbal compound Qiangji Yanghe Decoction acting on proteoglycan immunized BALB/c mice’s expression of serum tumor necrosis factor-α(TNF-α), matrix metalloproteinase-3 (MMP-3), the activation of nuclear factor receptor ligand (RANKL) and osteoprotegerin (OPG) and by pathological analysis, to explore the mechanism of strong ridge Yanghe Decoction treated on AS and bone destruction.Method:The choose of experiment animal were 32 rats aged 4-5 weeks old male BALB/c mice. After adaptive feeding a week,taking 24 of them modeling with proteoglycan and Freund’s adjuvant, the remaining 8 mice were normal group.Then modeling mice were randomly divide into three groups:model group, Qiangjiji Yanghe Decoction,SASP group,each group had eight mice.The Chinese medicine group was gavaged to strong ridge Yanghe Decoction(7.54g.kg-1),which connected with antlers,rehmannia, ephedra,guizbi,baijiezi and hive every day,SASP (0.02g.l0ml-1)was gavaged to SASP group,the normal group and model group were gavaged to normal saline.90 days after dosing in mice collected blood from orbit,and then obtain serum for TNF-α、MMP-3-. RANKL、OPG using Elisa method.Then mice were sacrificed by cervical dislocation for spine and joints analyzing by HE stain.Result:According to the expression of serum TNF-α、MMP-3、RANKL、OPG,found the expression of TNF-α、MMP-3、RANKL were high and OPG was low in model group,compared to normal group(P<0.05).The Chinese medicine group and SASP group set of serum TNF-a, MMP-3, RANKL levels were lower than the model ones, with significant distinguish (P<0.01 or P<0.05).But the two medicine group showed no significantly difference. The Qiangji Yanghe Decoction group and SASP group had higher level of OPG than model group(P<0.01 or P <0.05), results of two groups’comparison were the same as above.Pathological analysis showed Qiangji Yanghe Decoction can improve cartilage damageConclusion:1. Qiangji Yanghe Decoction possibly through reducing the expression of TNF-a and inhibit other inflammatory cytokine production, regulate immune response, reduce inflammation development.2.Qiangji Yanghe Decoction can increase the number of OPG,,reduce the expression of RANKL,thereby reduce the production of osteoclasts,by adjusting the RANKL system to inhibit the progress of bone destruction and achieve the effect of protection of bone.3.Qiangji Yanghe Decoction can reduce the expression of MMP-3,it may be reduce he activity of matrix metalloproteinases,,blocking multiple precursors, to slow down the process of bone estruction caused by cartilage degradation.
