| Objective:To study the expression of NUCB2 and Caspase-3 in benign prostatic hyperplasia(BPH),prostatic intraepithelial neoplasia(PIN) and prostate cancer(PCa). To explore the expression of NUCB2 and Caspase-3 in prostate cancer and its relationship. Analysis the correlation of NUCB2 and Caspase-3 in prostate cancer.Method:The expression of NUCB2 and Caspase-3 in benign prostatic hyperplasia tissue, prostatic intraepithelial neoplasia tissue and prostate cancer tissue was observed by the immunohistochemical SP method.Results:1.The positive expression rate of NUCB2 was 73.5% in PCa tissue. The positive expression rate of NUCB2 in PCa tissue was significantly higher than those in BPH, LGPIN, HGPIN tissue. And the positive expression of NUCB2 in BPH, LGPIN, HGPIN, PCa tissue were increased gradually, the difference was statistically significant(P <0.05).The positive expression rate of Caspase-3 was32.7% in PCa tissue. The positive expression rate of Caspase-3 in PCa tissue was significantly lower than those in BPH, LGPIN, HGPIN tissue. And the positive expression of Caspase-3 in BPH, LGPIN, HGPIN, PCa tissue were reduced gradually, the difference was statistically significant(P <0.05).2.In PCa tissue,the positive expression of NUCB2 was associated with Gleason grading and preoperative PSA(μg/L), the expression of NUCB2 in patients with survival of <5 years was higher than in patients with survival of ≥5years,the difference was statistically significant(P<0.05),the positive expression of NUCB2 was not related with the age of patients(P >0.05). The positive expression of Caspase-3 was not related with the age of patients,Gleason grading and preoperative PSA(μg/L).The positive expression of Caspase-3 in patients with survival of <5 years and patients with survival of ≥5 years were nosignificant differcece(P >0.05).3.The positive expression of NUCB2 in high differentiation of prostate cancer was significantly higher than that in HGPIN, the difference was statistically significant(P <0.05). The positive expression of Caspase-3 in high differentiation of prostate cancer and HGPIN were no significant differcece(P >0.05).4.The positive expression of NUCB2 and Caspase-3 in prostate cancer were negative relationship(r=-0.567,P =0.000).Conclusion:1.The positive expression of NUCB2 in PCa was significantly higher. It suggests that NUCB2 may participate in the occurrence of prostate cancer,and NUCB2 may become a useful target for prognostic in PCa.2.The positive expression of NUCB2 in high differentiation of prostate cancer was significantly higher than that in HGPIN.It suggests that NUCB2 may be used to differential diagnosis.3.The positive expression of NUCB2 and Caspase-3 in prostate cancer were negative relationship.It suggests that they may have a interrelated mechanism in the prostate cancer, combinded detection of NUCB2 with Caspase-3 may help the clinical diagnosis of prostate cancer. |
