Objective:Prostate cancer, prevalence rising rapidly, and in the majority of patients with advanced, loss surgery opportunity. In recent years, argon-helium cryoablation play an important role in the clinical treatment of prostate cancer. Mostly malignant invasive growth, larger, irregular, invades adjacent vital organs, leading to a variety of factors cryoablation may be incomplete, that the presence of residual tumor tissue phenomenon, the remaining parts of recurrence or metastasis is long-term One important factor in the efficacy. After the topical treatment of the biological characteristics of the residue tumor cell is changed? Turn on the body and what impact? In response to these problems, the study of mouse prostate cancer model constructed to observe the changes of lung metastases after cryotherapy, using immunohistochemistry, ELISA and other methods to analyze invasive treatment of residual tumor tissue after freezing, migration capability.Materials and Methods:1. The establishment of mouse prostate cancer were randomly divided into 2 groups:control group(untreated) and cryotherapy group, each with 40.2. Observe and record changes in tumor volume and survival of mice.3. Respectively, before cryosurgery, frozen after seven days, the mice were sacrificed14 days after freezing, obtain tumor tissue, lung and serum specimens. HE staining statistical incidence of lung metastases, immunohistochemical staining method to detect the expression of MMP-9 protein in tumor tissues and E-Cadherin in, ELISA serum MMP-9 content; and inter-group statistical analysis of these indicators at different time points change.Results:1. Compared with the control group, 7 days after freezing group, tumor volume at day14 was significantly reduced(P <0.05); compared with 7 days after surgery, freezing the tumor volume after 14 days no significant change(P> 0.05).2. Compared with control group, postoperative pulmonary metastasis rate freeze 7days no significant change(P> 0.05), after 14 days of pulmonary metastasis was significantly lower(P <0.05); lung metastasis rate in the control group of mice over time extend increased(P <0.05)), but the frozen lung metastasis rate little changed at all time points(P> 0.05).3. Frozen median survival time of mice was higher(25d vs.19d), with significant difference(P <0.05).4. with time, MMP 9-expression in tumor tissue in control group gradually increased,14 days increased significantly(P <0.05). 7 days before the expression of MMP-9group was significantly higher than the freezing technique(P<0.01), 14 天decreased significantly(P <0.01).5. Compared with control group, after freezing the serum MMP-9 were significantly increased after 7 days and 14 days in serum MMP-9 levels were higher than control group(P<0.05).6. with time, the E-Ca control group and frozen tumor tissue expression decreased(P<0.05); frozen after 7 days and 14 days E-Ca of expression levels were higher(P<0.05).Conclusions:1. Cryoablation effectively inhibit tumor growth and prolong the survival time of tumor-bearing mice.2. Compared to the control group, MMP-9 expression of local residual tumor tissue decreased after cryoablation, E-Ca expression increased, indicating reduced ability of invasion and metastasis, stable state, down mouse lung metastasis rate.
