Cortex Moutan is the dried root bark of Paeonia suffruticosa Andr.from Ranunculaceae, which is commonly used as traditional Chinese Medicine.It tastes bitter and acrid witht slightly cold,and it belongs to the heart, liver and kidney meridian. Cortex Moutan has the effect of clearing heat and cooling blood, promoting blood circulation and removing blood stasis.The peony bark has rich health care and medicinal value,it has a good curative effect on the aspect ofanti-bacterial,anti-inflammatory, protecting liver, reducing blood pressure,treating cardiovascular disease,anti myocardialischemia,anti coagulation, reducing blood sugar,anti blood fat,regulating of immune cells and inhibiting the central nervous system(CNS).In the Yuan dynasty, Cortex Moutan had been began to be charred and be used for clinical purposes.The long-term clinical practice proved that Cortex Moutan charcoal has obvious effect of cooling blood hemostatic,and it was used to treat the variety of hemorrhagic disease in clinic with definite curative.But now,no more detailed reports can prove that why it would have different effect for Cortex Moutan before and after being processed,which could change the effect of cooling blood and promoting blood circulation into cooling blood hemostatic.Related researchs found that the ethyl acetate fraction of Cortex Moutan charcoal is the main hemostatic active site. So being based on previous research work,we studied on the chemical constituents of the ethyl acetate part in order to make clear what type of chemicalcomponents in the ethyl acetate fraction, and to explore the hemostatic pharmacodynamic material basis execpt for carbon and tannin from CortexMoutan.Hoping to lay a material foundation for Cortex Moutan charcoal’s hemostatic efficacy study, and to provide a scientific foundation for the processing mechanism and hemostatic mechanism of Cortex Moutan charcoal.By using silica gel column chromatography, preparation type thin-layer chromatography(PTLC), MCI column chromatography, ODS reversed-phase column chromatography, Sephadex LH-20 column chromatography and recrystallization various separation techniques,22 compounds had been isolated from the ethyl acetate fraction of Cortex Moutan charcoal,and 17 compouds’ structures were identified by using NMR and EI-MS spectroscopic methods.These compounds include Paeonol(1),Catechol(2),3-hydroxy-4-methoxyacetophenone(3),benzoic-acid(4),2,5-dihydroxy-4-methylacetophenone(5),2,5-dihydroxy-4-methoxyacetophenone(6),methyl-linoleate(7),β-sitosterol(8),methyl-4-hydroxybenzoate(9),resacetophenone(10),2,3-dihydroxy-4-methoxyacetophenone(11),3-hydroxy-2-methyl-4-pyrone(12),3,8-dihydroxy-2-metylchromone(13),betulinic-acid(14),3β-hydroxy-olean-12-en(15),1-Monolinolein(16),and 2-(2,5-dihydroxy-4-methyl-phenyl) ethyl propionate(17). Among them,compound 17 is a new compound, compound 2,9,12,13,15 and 16 are isolated from Cortex Moutan charcoal.
