Pine leaves refer to acicular leaves of kinds of pinaceae plants, which are called pine needles,pine capills and so on. It were used for curing apoplexy, bone arthralgia and beriberi of migratory arthralgia in Traditional chinese medicine, the modern study found that the pine needles had pharmacological effect on antioxidant, antibacterial, antitumor, antivirus and so on.Pinaceae plants consisit of 10 genus and 113 species in China, which are widely distrubuted in China. Cedrus deodara is one of the pinacea, pine needles are a rich source, but so far have not been valid developed and utilized, In recent years the scholar study found that xylem of Cedrus deodara had antitumor active.In order to sduty more antitumor active ingredients and develope sufficiently the natural resources of pine needles of Cedrus deodara. First of all, this thesis summarized the domestic and foreign research results and reviewed on the chemical constituents and pharmacological activities about the botanical pine leaves. Secondly, we performed isolation and structure identification of the chemical constituents of ethyl acetate fraction from pine needles of Cedrus deodara.24 compounds were isolated from the 95% EtOH extract by means of silica gel and sephadex LH-20 column chromatography etc. The structures were elucidated on the basis of chemical evidence, extensive spectroscopic analysis (1H-NMR,13C-NMR,DEPT,HR-ESI-MS,HMBC) and the literature data. The 19 compounds were identified as myricetin (1),myricetin-3-β-D-glucopyranoside(2),3',4'-di-methylmyricetin-3-O-β-D-glucopyranoside(3),myricetin-3-O-(6"-O-E-coumaroyl)-a-D-glucopyranoside(4),3',5'-di-methylmyricetin-3-O-(6"-O-acetyl)-a-D-glucopyranoside(5),isorhamnetin-3-O-β-D-glucopyranoside(6),dihydromyricetin(7),cedrin(8),quercetin(9),dihydroquercetin(10),hyperoside(11),quercetin-3-O-(3",6"-O-E-p-coumaroyl)-β-D-glucopyranoside(12),kaempferol(13),kaempferol-3-O-β-D-glucopyranoside(14),kaemp ferol-3-O-(6"-O-E-feruloyl)-β-D-glucopyranoside(15),kaempferol-3-O-(6"-O-E-coumaroyl)-β-D-glucopyranoside(16),shikimic acid (17),shikimic acid butyl ester(18),protocatechuic acid (19), Compounds 4 and 5 are new compounds, except for compounds 17 and 19, other compounds are isolated from pine needles of this genus for the first time. Finally, we established a method that is simultaneous determination for contents of myricetin, quercetin and kaempferol from the pine needles of Cedrus deodara by HPLC. The results showed that the method is simple, rapid and accurate, the contents of myricetin, quercetin and kaempferol are separately 0.185%,0.422%,0.751%.The thesis enriched natural compounds data base of the pinaceae by studying on ethyl acetate fraction from pine needles of Cedrus deodara, the scientific basis is provided for the further development of pine needles of Cedrus deodara.
