| Objective:On the basis of gansu Chinese dangshen, astragalus membranaceus different formula, for building successful precancerous lesions of chronic atrophic gastritis(PLGC)model rats intervention treatment.Precancerous lesion of chronic atrophic gastritis(PLGC)molecular biology index CDX2, Survivin and the influence of the expression of PTEN and to study the effect of different TCM syndrome differentiation, reagent dosage for clinical legislation, improve the curative effect.Methods:Will 68 Wistar rats(male and female half) random separated 16(male and female half) as the blank group, separated only 52(male and female half) to build module;Blank group(conventional breeding), made the module to 20 tendency/L deoxycholic acid sodium solution 2 ml daily on an empty stomach lavage(the fast one hour) before and after the lavage, drink 0.05% of ammonia, combined with the hunger disorder(two-day optional one-day fast casual water, drink enough feeding), make mould with above method.In building 10 weeks and 12 weekends in blank group and module of each two rats, stomach again after HE staining examination, on the basis of chronic atrophic gastritis(the CAG)pathology judgment model success, 12 weeks, the decision made after the success of the module, module in the rats of survival is to be built and randomly divided into four groups,namely model group, yiqi spleen and replenishing qi spleen dry wet group and yiqi spleen blood group(12), model group give saline(2 ml/only), replenishing qi spleen and replenishing qi spleen dry wet group and yiqi spleen blood group respectively for crude drugs containing 2.7 g/kg, 5.4 g/kg, 5.4 g/kg medicine mixed suspension 2 ml lavage, once a day, to fill the stomach full 12 weeks.Blank group rats(12), the conventional breeding, test all executed after 24 weeks, materials processing.Then observed the pathological changes of rat gastric mucosa, using immunohistochemical method to detect CDX2, Survivin and the expression of PTEN.Please observe, pathologist CDX2, Survivin and PTEN gene expression for the particles in the cytoplasm and nucleus tan is positive particles, positive decision criteria: each section randomly selected five high vision(20 x20) detection, positive cells accounted for more than 10% positive expression, < 10% negative.Results:(1) Qi spleen and replenishing qi spleen and replenishing qi and invigorating blood circulation of the different treatment can inhibit CDX2 expression, inhibition of expression of CDX2 replenishing qi of the spleen blood group was obviously better than the other two treatment groups(P < 0.05).(2) Qi spleen and replenishing qi spleen dry wet, replenishing qi invigorating blood circulation to different treatment can inhibit expression of Survivin, inhibit expression of Survivin replenishing qi of the spleen blood group was obviously better than the other two treatment groups(P < 0.05).(3) Qi spleen and replenishing qi spleen dry wet, yiqi the invigorating blood circulation to different treatment can increase the expression of PTEN, yiqi raise PTEN expression of spleen blood group was obviously better than other two treatment group(P < 0.05).Conclusion : Qi spleen and replenishing qi spleen dry the different treatment of traditional Chinese medicine for replenishing qi and invigorating blood circulation, which can effectively improve the stomach precancerous lesion rat gastric mucosa pathological condition, cut CDX2 and Survivin expression, raise level of PTEN gene, which the most notable curative effect, replenishing qi of the spleen blood group that is given priority to with replenishing qi spleen of different treatment of traditional Chinese medicine can be effective intervention on gastric precancerous lesions, replenishing qi invigorating blood to intervention is more apparent. |
