Influence Of Traditional Chinese Medicine Decoction-Fu Wei Jian To Precancerous Lesions Of Gastric Cancer In Rat And Its Relevant Factors

Objective This experimental research studied the influence of Fu wei jian to Pre cancerous lesions of gastric cancer in rat and its relevant factors including VEGF、CO X-2、To evaluated the therapeutic action about it,the molecular biology mechanism of influence about fu wei jian decoction to Precancerous lesions of gastric cancer in rat is disscussed.Methods 1.Randomly chosen 20 as normal rat group out of 90 male wistar rats weight 200～240g.The remaining 70 rats were established animal models.2.After the model been successfully built,rats randomly divided into four group:model control gr oup,Fu Wei Jian Decoction high dose treatment group,Fu Wei Jian Decoction middl e dose treatment group and Fu Wei Jian Decoction low dose treatment group.And th e rats in normal rat group randomly divided into two group:normal control group and Chinese medicine control group,each group with 10 rats.3.Rats’ stomachs of Chin ese medicine treatment group were irrigated with high,middle or low doses of Fu We i Jian Decoction in eight weeks.The rats in Chinese medicine control group were irri gated with Fu Wei Jian Decoction in middle dose.And the rats in normal control gro up or model control group were irrigated with NS.4.The expressions of COX-2 and VEGF in gastric mucosa were analyzed by immunohistochemistry and RT-qPCR.Results 1.The levels of COX-2 and VEGF both rose in gastric mucosa of rats in model control group.2.Compared with model control group,the expressions of CO X-2、VEGF and their mRNA in gastric mucosa of the three groups irrigated with F u Wei Jian Decoction were reduced(P<0.05)。But the expressions of Chinese medic ine control group declined without statistical significance compared with model control groups(P>0.05)。3.The expressions of VEGF and its mRNA of Fu Wei Jian Dec oction high dose treatment group were lower than Fu Wei Jian Decoction middle dose treatment group(P<0.01)and Fu Wei Jian Decoction low dose treatment group(P<0.05).Conclusion 1.Fu Wei Jian Decoction could decline the expressions of COX-2 an d its mRNA in rat’s gastric mucosa with precancerous lesions of gastric cancer.2.Fu Wei Jian Decoction may promoting the expression of COX-2 in gastric mucosa with heath rats.3.Fu Wei Jian Decoction could decline the expressions of VEGF and its mRNA in rat’s gastric mucosa with precancerous lesions of gastric cancer,maybe disp lay certain dosage-effect relationship.
