Objective: To detect the effects of Compound Malt Pills on weight changes, level of T and INS in serum, expressions of ERα, ERβ, AR and INSR. To explore the target and mechanism of Compound Malt Pills on treating PCOS and lay a clinical foundation.Method: 48 female Sprague-Dawley rats aged 6 weeks were randomly divided into Normal group(n=8) and rat model group(n=40). 0.125 mg·mL-1 letrozole-CMC solution was used for 21 days lavage to build a PCOS rat model. The successful modeling rats were randomly divided into Model group(saline), Diane-35 group(1.44 g · kg-1), Low dose group(5.76g·kg-1), Medium dose group(11.52g·kg-1) and High dose Compound Malt Pills group(23.04g·kg-1), each group was treated by corresponding drug for lavage 21 days after successful modeling. And Normal group was lavaged 21 days by saline. Weight changes were detected every day from beginning. Ovarian morphology changes were observed by HE staining, level of T and INS in serum were measured by ELISA, and expressions of ERα, ERβ, AR and INSR were examined by immunohistochemistry, Western blot, and Real-time PCR respectively after treatment.Results:(1)After successful modeling, weight of each groups increased comparing with Normal group(P<0.01), but did not change significantly comparing with Model group after treatment(P>0.05).(2)The number of cystic follic les in Model group increased and granulosa cells decreased. Comparing with Model group, the number of cystic follic les reduced, but corpus luteum and granulosa cells increased.(3)Comparing with Normal group, level of serum T, INS in Model group increased significantly(P<0.05 or P<0.01), and decreased in Compound Malt Pills groups comparing with Model group(P<0.05 or P<0.01).(4)Expressions of ERα and ERβ in Model group significantly decreased(P<0.01), but the AR and INSR increased(P<0.01). Comparing the intervention groups with Model group, differences of ERα, ERβ in Compound Malt Pills groups increased(P<0.05 or P<0.01), but expressions of AR and INSR decreased(P<0.01).Conclusion: Compound Malt Pills may correct the endocrine metabolic disorder in PCOS model by improving ovarian morphology, reducing level of T and INS in serum, upregulating the expressions of ERα and ERβ and downregulating the expressions of AR and INSR.
