Objectives To investigate the effect of Yangxue Q ingnao granules on the expression of Glial cells proliferation in CA1 area and neural precursor cells in dentate gyrus of hippocampus in rats with vascular dementia, to study regulatory role of Yang Xue Q ing Nao granules on controling inflammation and Promoting neurogenesis in vascular dementia rats.Methods 144 Sprague-Dawley rats were randomly divided into sham operation group(n=48),VD group(model group, n=48) and Yangxue Q ingnao granules treatment group(treatment group, n=48).The VD model group was prepared by modified Pulsineli’s fourvessel occlusion.The sham operation group and the model group were given normal saline 10 ml/(kg/d) by gavage,and the treatment group was given Yangxue Q ingnao granule 3.2 g/(kg /d) by gavage.The learning and memory abilities of rats were detected by the Morris water maze.In C A1 area of hippocampus,the expression of GFAP and CD11 b was observed by immunohistochemical analysis and Western blotting method 1, 2, 4 and 8 weeks after modeling.The expression of b FGF in CA1 area of hippocampus were detected by Immunohistochemistry at 1, 2,4 and 8 weeks after vascular dementia rat model operation.The expression of Nestin was detected with Western blotting,the expression of 5-bromodeoxyuridine(Brd U) and Brd U/Nestin were detected with immunofluorescence in dentate gyrus of hippocampus 1, 2, 4 and 8 weeks after modeling.Obeserve tissue section with Microscope camera,and then analysis and statistics.Results 1 Compared with the model group,the learning and memory abilities of rats in the treatment group were significantly improved in 3-5 day.2 The expression of GFAP in CA1 area of hippocampus was significantly higher in the model group than in the sham operation group(P<0.01),and was lower in the treatment group than in the model group(P<0.01) at every time point.3 Compared with sham group,the expression of CD11 b in CA1 area of hippocampus of vascular dementia rats were significantly increased in model group at different time points(P<0.01);Compared with model group,the expression of CD11 b in CA1 area of hippocampus of vascular dementia rats were significantly decreased in treated group at different time points(P<0.01),especially after 2 weeks.4 The protein expressions of b FGF were detected by the immunohistochemical analysis.The results showed that compared with the sham operation group,there was obvious learning and memory disorders in the model group with increased protein expressions of b FGF in CA1 area of hippocampus at difference time points(P<0.01).5 The expression of Nestin,Brd U and Brd U/Nestin increased in the model and treatment groups with time,peaked at 4 weeks after modeling,and it was more than that of the sham group on all the time points(P<0.01).However, it was more in the treatment group than in the model group on all the time points(P<0.01).The expression was the highest on the 4th week.Conclusions It was concluded that Yangxue Q ingnao granules could inhibit the activation and proliferation of astrocytes and microglia in CA1 area of hippocampus,then promote the proliferation of neural precursor cells in dentate gyrus and increase the expression of b FGF hippocampus in vascular dementia rats,by which way YXQN Granules may improve the learning and memory.
