Objectives Using microarray technology, screening characteristic circRNA expression profile of lung adenocarcinoma, and then exploring the role of these circRNA in gene regulation, meanwhile establishment target circRNA of further clinical verification in lung adenocarcinoma. The aim is screen new types of molecular markers and molecular target for therapy in lung adenocarcinoma.Methods Paired collection surgical removal samples of carcinoma tissue and noncarcinoma adjacent to carcinoma tissues of 6 lung adenocarcinoma patient. AfterRNA extraction, reverse transcription, testing by microarray, then scan images. After all above computer processing the data through the analysis of the software, combining with gene library resources, probe into the regulation mechanism of circRNA.In the end, compring each other for all circRNA,deterrmine the circRNA for clinical vertifing in the next study.Results 1 There is different in circRNA Expression profile for lung adenocarcinoma tissue with adjoining non-carcinoma. Compared with the non-carcinoma tissue adjacent to carcinoma, lung adenocarcinoma tissue up express circRNA 595 in probe CBC1, down express circRNA 41. Two probe at the same time, up expression circRNA105, down expression of 11.(p ≤5%, and FC(abs) > 2.0). 2 Using the known metabolic disorders and diseases related signaling pathways database and database, annotation for each circRNA in pathways, disease and functional. 3 Hsacirc0000576 is a typical circRNA in lung adenocarcinoma.It is the candidate for the next clinical study.Conclusions 1 The circRNA microarray screening for lung adenocarcinoma got characteristic circRNA expression profile, up expressions 105, down expression 11. 2 Combined with the existing gene library resources, and by use of KOBAS software analysis, confirmed the lung adenocarcinoma circRNA expression profile, then for each circRNA, comments and annotation in pathways, disease and effects of the miRNA targets, and analyzed the characteristics of lung adenocarcinoma circRNA significant enrichment of pathways, significant enrichment of disease, and significant enrichment of Gene Ontology entries. Meanwhile simulates the process of biology, cell and molecular components function three aspects of Gene Ontology functional hierarchical network diagram. 3 The study established hsacirc0000576 as a target circRNA verification in 66 lung adenocarcinoma patients in clinical, and to explore the correlation of circRNA in age, gender, smoking status, and different clinical features such as T and N staging.
