Objective:To study the effects of flurbiprofen axetil and dezocine combined analgesia to wound healing and the relationship with GM-CSF, so as to provide references for clinical medication.Methods:160 healthy male Kunming mice were selected randomly, which were divided into:normal saline group(NS group); dezocine group(group D); flurbiprofen axetil group(group F) and dezocine +flurbiprofen axetil group(D+F group), each group included forty mice. group F, group D, group D+F was respectively injected by intraperitoneal injection of dezocine 5 mg/kg, flurbiprofen axetil 25mg/kg, dezocine 5 mg/kg + flurbiprofen axetil25mg/kg 10 min before skin incision. The allowance was filled with equal volume saline.Ten mice were selected from each group in 3, 7, 10 days to record the wound healing rate. Take the skin tissue around the wound and detect the content of GM-CSF by ELISA method, meanwhile, detect the protein content by BCA method, last, the ratio of the content of GM-CSF and protein content could use to represents the concentration of GM-CSF.Selecte wound tissues from 3 days and 7 days to make pathological slices.Observe the wound pathological changes of the rest 10 mice from each group to calculate the time of wound healing.Results:Results of various data indicators: D + F group wound healing time(11.02±0.81) d, 7days wound healing rate(78.765±1.972) %, 10 days wound healing rate(87.970 ±2.281) %were higher than the other three groups, the difference was statistically significant(P<0.05), 3 days wound healing rate(26.394±0.835) % compared with group D(27.741±1.538) %, there was no statistically significant difference(P > 0.05), group D+F 3, 7, 10 days of GM-CSF concentration(23.252±1.628) %,(43.898 ±2.285) %,(35.861±2.739) %were higher than that of NS group and F group respectively, lower than that of group D,the difference was statistically significant(P<0.05). The 3 day wound healing rate(19.245±1.331) of group F is less than the NS group(20.439+0.890), the difference has statistical significance, and the wound healing rate(67.955±2.322) of 7 days, the wound healing rate(80.955±1.773) of 10 days wer respectively greater than NS group of seven days the wound healing rate(62.970±157) and the wound healing rate of(79.056±178)10days, GM-CSF concentrations were lower than which is in NS group.Pathologic findings: 3 days after trauma, only a small amount of fibroblasts can be see in NS group, and a certain number inflammatory cells infiltration, F group of inflammatory cells show less than group NS, group D has a small amount of inflammatory cells infiltration and fibroblast, D+F group has a small amount of fibroblast cells and inflammatory cells; 7 days after trauma:NS group of inflammatory cells show more, and group D has less fibrous cells, blood capillary is scarce, group Dand group F have more collagen fibers, capillary, squamous epithelial thickening, D+F group, has a large number of fibroblasts, collagen fiber, less inflammatory cell.Conclusion:25mg/kg flurbiprofen axetil and 5mg/kg dezocine both can promote wound healing,but flurbiprofen axetil for early wound healing will have different degree of delay, and assist dezocine combination can mitigate the inhibition which can also greatly promote healing of the wound forward at the same time, dezocine has promoting effect to the GM-CSF, which flurbiprofen axetil has inhibitory effect on it.
