Objective To investigate the experssions of Versican and VEGF in gallbladder carcinoma and their correlations with angiogenesis.Methods The expressions of Versican,VEGF and CD34 in 72 cases of gallbladder carcinoma tissues(tumor group),40 cases of Gallbladder intraepithelial neoplasia tissues(GBIN group),30 cases of chronic cholecystitis tissues(inflammatory group) and 20 cases of normal gallbladder tissues(normal group) were detected by immunohistochemistry staining, and the relations of Versican,VEGF and CD34 expressions with clinicopathologic variables(age,gender, TNM stage, differentiation degree, lymph node metastasis) and prognosis of gallbladder carcinoma were analyzed. all of tissues were resected at the 101 th Hospital of PLA from 2008-1 to 2015-1. Application SPSS16 statistical software analysis experimental results, Using chi-square test analysis count data, Using t test analysis two samples, multiple sample were compared by ANOVA, Using the Kaplan-Meier method analysis survival curves, and test by Log-rank, correlation analysis using Spearman rank correlation analysis. Test standard α = 0.05.Results The expression of Versican was signifcantly higher in tumor group[66.7%(48/72)] than that in normal group[5.0%(1/20),?(17)(28) 23.910(11) P<0.05],the inflammatory group [36.7%(11/30),?(17)(28)7.816(11) P<0.05] or GBIN group [47.5%(19/21),?(17)(28)3.930,P<0.05].The expression of Versican was signifcantly higher in GBIN group and inflammatory group than that in normal group(?(17)=10.838;?(17)=6.597,both P<0.05), but the statistically significant difference was not found between GBIN group and inflammatory group(?(17)=0.822,P>0.05).The expression of VEGF was higher in tumor group[73.6%(53/72)] than that in normal group[15.0%(3/20),?(17)(28) 22.575(11) P<0.05],the inflammatory group [16.67%(5/30),?(17)(28)27.995(11) P<0.05] or GBIN group [45.0%(18/40),?(17)(28)9.071,P<0.05]. The expression of VEGF was signifcantly higher in GBIN group than that in normal group(?(17)=5.275,P<0.05) and inflammatory group(?(17)=6.238(11) P<0.05),but the statistically significant difference was not found between inflammatory group and normal group(?(17)=0.000,P>0.05).The average MVD of tumor group(48.08±13.36) was signifcantly higher that in normal group [(25.65±7.77), t=9.563,P<0.05],the inflammatory group [(26.97±7.31),t=10.227,P<0.05 ]or GBIN group [(36.45±8.95), t=5.192,P<0.05]. The average MVD of GBIN group was higher than that in normal group(t=4.934,P<0.05) and inflammatory group(t=4.728(11) P<0.05), but the statistically significant difference was not found between inflammatory group and normal group(t=0.608,P>0.05). The expressions of Versican,VEGF and MVD were closely related to the lymph node metastasis and TNM stage of gallbladder carcinoma(all P<0.05), moreover, the expressions of Versican and MVD were also relevant to the differentiation degree of gallbladder carcinoma. Survival analysis showed that the survival rate in patients with positive Versican or VEGF expression was signifcantly lower than that in patients with negative Versican or VEGF expression(?(17)(28)(24)(13)(17)(15)(20)(11) ?(17)(28)(21)(13)(21)(15)(17),both P<0.05),the low expression of MVD had a significantly higher survival time than in the high expression group(?(17)(28)(20)(13)(16)(23)(16),P<0.05),. There was a positive correlation between the expressions of Versican and VEGF in gallbladder carcinoma tissue(r=0.513,P<0.05),Moreover, the expression of MVD in gallbladder carcinoma tissue had a positive correlation with Versican and VEGF respectively(r= 0.402, r=0.437, both P<0.05).Conclusion Versican and VEGF probably participates in the process of occurrence and development of gallbladder carcinoma and were closely associated with tumor angiogenesis.and positive expression of Versican and VEGF predicts poor prognosis of gallbladder carcinoma patients.
