Objective To investigate the relationships between the gene overexpression of PDSS2 and the biological behavior(Cell growth, cell proliferation, cell cycle,apoptosis, cell migration and invasion) of human colon cancer, provide the basis for a clear relationship PDSS2 with colon cancerMethods 1、Construct the PDSS2 over-expression vector;2、To transfect SW480、HCT116 cells;3、Set the empty vector transfected control group(SW480、HCT116)、PDSS2 over-expression plasmid in the experimental group(PDSS2);3、The MTSwas used to evaluate cell growth and proliferation.;4、The flow cytometry was employed to analyze the cell cycle and apoptosis rate of SW480、HCT116cells in vitro induced by PDSS2;5、The cell migration assay and the cell invasion assay were performed in Transwells and scratch assay.Results(1) a、The d1、d2 and d3 proliferation rates the experimental group were117.54%、161.19% and 316.79%. And proliferation rates of the SW480 group were127.24%、206.72% and 425.00%. There were statistically significant difference(P<0.01) at each time point. The d1、d2 and d3 inhibition rates of PDSS2 overexpression on SW480 cell proliferation were 7.62%、22.02% and 25.46%, at the same time points were 0.00% 、 0.00% and 0.00% in the SW480 group. There were statistically significant difference(P <0.01) at each time point.b、The d1、d2 and d3 proliferation rates the experimental group were 115.04%、150.81% and 248.37%, And proliferation rates of the HCT116 group were118.97%、164.43% and 289.33%. There were statistically significant difference(P<0.01) at each time point. The d1、d2 and d3 inhibition rates of PDSS2 overexpression on HCT116 cell proliferation were 5.98%、10.82% and 16.53%,, at the same time points were 0.00%、0.00% and 0.00% in the HCT116 group. There were statistically significant difference(P <0.01) at each time point.(2)a、In the experimental group, G1 phase cells accounted for 70.24%, 59.54% in the SW480 group. S phase cells accounted for 25.00% in experimental group, lower than the 33.86% in the SW480 group. G2 phase cells accounted for 4.77%, 6.60% in the SW480 group.b 、 In the experimental group, G1 phase cells accounted for 68.25%, 57.10%in the HCT116 group. S phase cells accounted for 18.12% in experimental group,lower than the 24.14% in the HCT116 group. G2 phase cells accounted for 13.63%,18.76% in the HCT116 group.(3)a、 Total apoptotic cells in the experimental group was 11.03% and the SW480 group was7.58%(P=0.012).b、 Total apoptotic cells in the experimental group was 16.03% and the HCT116 group was 13.01%(P=0.015).(4)Scratch assay showed distance between cells on SW480-PDSS2 group is |445.62±27.45|,the control group is |335.03±31.69| after 48 h.;distance between cells on HCT116-PDSS2 group is |259.34±13.2|,the control group is|188.38±9.5|| after 48 h..(5)a、In transwell assay, the number of migrating cells in the experimental group was136.5±13.25 and the SW480 group was 350.50±30.17. The gaps were statistically significant(P <0.01). In experimental group, the number of invasive cells was 59.17±9.85 corresponding to the SW480 group was 140.50±27.08. There was statistically significant difference(P <0.01).b、In transwell assay, the number of migrating cells in the experimental group was19.83±2.32 and the HCT116 group was 63.17±16.07. The gaps were statistically significant(P <0.01). In experimental group, the number of invasive cells was8.83±1.72 corresponding to the HCT116 group was 37.33±4.27. There was statistically significant difference(P <0.01).Conclusions(1) PDSS2 overexpression can inhibit the proliferation of SW480、HCT116 cells, preventing their transition from G1 to S phase.(2) The overexpression PDSS2 can affect the regulation of SW480、HCT116 cell cycle.(3) PDSS2 overexpression can induce apoptosis of SW480、HCT116 cells.(4) The overexpression PDSS2 can prevents the migration and invasion of SW480、HCT116 cells.
