Objective:This paper is aim to disclose the relation existing between uterine injection of OPN and embryonic implantation, to investigate the safety of ntrauterine injection of OPN from endometrial cancer occurrence and development, and to improve the rate implantation rate of ART.Methods:(1) This paper used 6-week-old sexual maturity ICR mice for the study, the use of intrauterine injection method was different from the classical surgical intrauterine injection, which one lcm long incision was made on the abdominal midline above the edge of the pubic symphysis 0.5 cm, tried to simplify and improve the classic surgical intrauterine injection;(2) the OPN elisa kit was used for detection of uterine OPN expression level before and after injection, to evaluate the efficiency of modified intrauterine injection;(3) I established the natural conception model of mice, and according to the different interventions, the ICR mice were randomly divided into three groups:control group, NS group, OPN group (n=8), on the fourth day after pregnancy (D4) the mice were injected of OPN or NS by improved surgical intrauterine injection, and on the eighth day after conception (D8) the mice were sacrificed, the pregnancy rate and the number of implantation was observed to explore the function of OPN during the implantation in mice;(4)1 detected endometrial E-cad and VIM expression by immunohistochemistry after intrauterine injection, to explore the effects of OPN injection on endometrial cells EMT, to investigate the safety of intrauterine injection of OPN.Results:(1) OPN expression levels in NS group declined slightly (9.91±5.75ng/L) compare to the control group (16.06±3.96ng/L) but the difference was not statistically significant (P>0.05), OPN group OPN expression levels (60.20±15.10ng/L), significantly higher than the NS group and control group, statistically significant difference (p<0.05).(2) The pregnancy rate and the average number of implantation(12.5%,0.3±0.7) in NS group are below control group (75.0%,9.1±5.9), and OPN group (62.5%,4.6± 4.1), this differences has statistics meaning (p<0.05); The pregnancy rate and the average number of implantation in the OPN group found below control group, but the differences in pregnancy rate has no statistics meaning (p>0.05), differences in the average number of implantation found to have statistics meaning (p<0.05);(3) E-cad strong positive (+++) rate expressions was 75%, VIM weakly positive (+) expression was 75%, in the control group, NS group, and OPN group, there was no significant differences found between the three (p>0.05).Conclusions:(1) Improving OPN uterine injection procedure, can improve uterine OPN level of expression;(2) Improving the intrauterine procedures of NS can significantly reduce the pregnancy and implantation rate among mice;(3) OPN intrauterine injection can improve the pregnancy rate and the embryonic implantation in mice;4) OPN intrauterine injection had no effect on mice endometrium EMT.
