Objective: To observe radix isatidis’ induction effect on oral epithelium RBD-2 gene expression in rats, to explore the ideal effect relationship between drug concentration and treatment time, and to lay a foundation of the research on therapeutic effect of radix isatidis on oral disease models in rats. Methods: 135 Wistar rats were randomly divided into 5 groups: Group A(positive control group), Group B(blank control group), Group C(high-dose group), Group D(middle-dose group), and Group E(low-dose group), and each group has 27 rats.Group A(positive control group): 10 mg lipopolysaccharide of coli bacillus(LPS) was dissolved in 100 ml physiological saline, and 2 mg/kg LPS was given as stimulant on alternate-days;Group B(blank control group): regular feeding, with no irritants;Group C(high-dose group): radix isatidis injection was given daily at 3.56ml/kg through tail intravenous injection;Group D(middle-dose group): radix isatidis injection was given daily at 1.78ml/kg through tail intravenous injection;Group E(low-dose group): radix isatidis injection was given daily at 0.89 ml/kg through tail intravenous injection; On Day 3, Day 5, Day 7 of the trial, 9 rats from each group were sacrificed, and the gingival tissues were taken, labeled and frozen in liquid nitrogen. With r BD-2 as a primer, and β-actin(β-myosin) as an internal primer, r BD-2 gene expression level in the gingival tissues of rats was determined by Real time PCR technique. Results:1. Radix isatidis’ induction effect on oral epithelium r BD-2 gene expression in rats in the high-dose group is significantly higher than that of the middle-dose group and the low-dose group(P < 0.05), and there is no significant difference(P > 0.05) when compared to the positive control group;2. r BD- 2 gene expression level in the gingival tissues of rats increased with the extension of treatment time(P < 0.05). Conclusion:1. Radix isatidis has induction effect on r BD-2 gene expression in gingival epithelium of rats, and the expression level increases with drug concentration and treatment time, demonstrating an increasing tendency.2. Giving a high dose(2 times) of radix isatidis injection(3.56ml/kg) consecutively for 7 days, is safe and reliable, with significant induction effect, which can be taken as the reference basis for the research on therapeutic effect of radix isatidis on oral disease models in rats. |
