| Objective: Cisplatin(DDP) is the main chemotherapy drugs for ovarian cancer. But ovarian cancer cells tend to appear on cisplatin resistance in the clinical application. The tumor cells are sensitive to low dose irradiation. Low dose radiation therapy can improve the effect of chemotherapy drugs on ovarian cancer, but its mechanism is not clear. This topic will explored the mechanisms of low dose irradiation on ovarian cancer cells which are resistant to cisplatin(SKOV3/DDP) via survivin and capase-3.Methods: Cells were randomly divided into three groups, low-dose-radiation group(LDR),control group and conventional-dose group.Concentrations of cisplatin were 0, 0.625,1.25,2.5, 5and 10ug/ml.The LDR group was expousured to 0.5Gy irradiation for 0,8,16,24 h, and total dose was 2Gy. The conventional irradiation group was treated with 2Gy irradiation. The conventional group without irradiation.Cell viability was examined by cell counting kit-8(CCK-8) assay, Quantitative real-time PCR was used to detect Caspase-3 and Survivin expression level. Flow cytometery were used for detecting cell apoptosis.Results: The cell viability of LDR was lower than the control group and conventional irradiation group(P<0.05).The IC50 of LDR, control and conventional-dose group was 3.837±0.16,9.467±0.17 and 9.389±0.17.The cell apoptosis rate of LDR was higher than the other two groups.The cell apoptosis rateof LDR, control and conventional-dose group was,51.40%± 0.24,20.70% ± 0.59,39.20%± 0.29. The expression level of Caspase-3 m RNA was higher, Survivin m RNA was lower(P<0.05).Conclusion:The expression level of Survivin in ovarian cancer cells was higher than that of normal ovarian cells. Caspase-3 was lower in ovarian cancer cells.LDR may have the effect on reversing the ovarian cancer resistance through suppressing the Survivin expression level, improving Caspase-3 expression and promoting the SKOV3/DDP cells apoptosis.That may provide new methods for the clinical treatment of ovarian cancer... |
