Objective To investigate the effect of recombinant human endostatin on the expressions of VEGF and CD34 in a gerbil model of hepatic alveolar echinococcsis.Methods Fifty healthy female gerbils were randomly divided into five groups: model group(group A),saline control group(group B),low dose of Endostar injection group(group C),medium dosage of Endostar injection group(group D),Endostar injection group(group E).alveolar echinococcsis sp.was injected into abdominal cavity of the fifty gerbils to creat an animal model of hepatic alveolar echinococcosis.Each gerbil was injected with 0.1 m L of a suspension of immature suspension of an alveolar echinococcus sp.Thirty days after injection,respectively,Group C,D and E were given intraperitoneal injection in 3 courses(each course lasted 21days: 5/10/15 mg/kg Endostar on day 1-14 and respite for 7 days).Group B received a corresponding volume of saline in the same manner as Endostar group,Group A received no further treatment after injection.The gerbils were sacrificed once the treatment was finished,take the blood from their body and weigh cyst wet weight.The expressions of VEGF and MVD-CD34 were detected by enzyme-linked immunosorbent assay(Elisa)analysis,immunohistochemistry staining and Western Blot.Results Alveococcus tissues wet weight of Endostar group decreased,moderate dose and high dose of Endostar group had statistical significance in cyse wet weight(P<0.05).The expressions of VEGF and CD34 reduced in serum with the increase of Endostar,in addition to the model group and saline control group,and moderate dose and high dose of Endostar group,the VEGF expressions of other groups were statistically significant(P<0.05).There was significant difference between moderate dose of Endostar group and model group,as well as moderate dose of Endostar group and saline control group,high dose of Endostar group was the same as moderate dose of Endostar group(P<0.05).there was no significant difference between other groups(P<0.05)in the CD34 expression in serum.VEGF and CD34 were detected in Each gerbils' Alveococcus tissues,the positive cells were yellow or brown yellow granules,located in the cytoplasmic,also found in the cell membrane;the moderate dose and high dose of Endostar groups' VEGF immunohistochemical scores were lower than those in model group and saline control group,the difference was statistically significant(P<0.05),there was no significant difference among the low-dose of Endostar group,the model group,saline control group(P>0.05);Compared with model group and saline control group,MVD value OF each Endostar group decreased with statistically significant difference(P<0.05),there was no significant difference among each Endostar groups,between model group and saline control group(P>0.05).In Western blot test,VEGF and CD34 protein content also decreased,in addition to the model group and saline control group,and moderate dose and high dose of Endostar group,the VEGF expressions of other groups were statistically significant(P<0.05).The CD34 expression quantity of moderate dose and high dose of Endostar groups were lower than those in other groups,the differences were statistically significant(P<0.05),there was no significant difference between other groups(P>0.05).Conclusion Recombinant human endostatin can decrease the expression of vascular endothelial growth factor(VEGF)and microvascular density,and inhibit the growth ofEchinococcus multilocularis to a certain extent.
