| Objective: To investigate the wogonin in enhancing the sensitivity to cisplatin on cisplatin(CDDP)-resistance of human ovarian cancer SKOV3/CDDP cells, and to explore its possible mechanism.Methods: The resistance index(RI) of SKOV3/CDDP was detected by CCK-8 method, the migration and invasion abilities of SKOV3 and SKOV3/CDDP cells were detected by scratch wound healing and Transwell migration and invasion assay,respectively. The expressions of P-glycoprotein(P-gp), E-cadherin(E-cad), N-cadherin(N-cad) and vimentin(vim) was detected by Western blot(WB). Then SKOV3/CDDP cells were treated with 20 μmol/L Wogoninfor 24 h.The experimentwas divided into three groups that included SKOV3 cells group, SKOV3/CDDP cells group and SKOV3/CDDP +Wogonin cells group. The SKOV3 cells and the SKOV3/CDDP cells withoutany treatment were used as the controls. The cell proliferation abilitywas detected by CCK-8 method. The migration and invasion abilitieswere detected by scratch wound healing and Transwell migration and invasion assay, respectively. Furthermore, the expressions of P-gp, E-cad, N-cad and vim proteins were detected by WB.Results: The RI of SKOV3/CDDP was 5.6,the migration and invasion abilities of SKOV3/CDDP cells were significantlyincrease, the expressions of N-cad and Vim proteins were up-regulated, but the expression of E-cad was down-regulated. After treatment with Wogonin for 24 h, the sensitivity of SKOV3/CDDP cells to CDDP was increased, and the migration and invasion abilities were significantlydecreased; the expressions of N-cadand Vim proteins weredown-regulated, and the expression of E-cadwas up-regulated, showed significant differences in different group. While there was no significant difference in the expression of P-gp among differentgroups.Conclusion: Wogonin may increase the sensitivity of SKOV3/CDDP cells to CDDP by reversingepithelial-mesenchymal transition(EMT). |
