Objectives Through anti-inflammatory、 antioxidant sress and other biological effect,Rosuvastatin can improves the survival rate of the transplanted bone marrow stromal cells(SMSCs).The further purpose of this experiment is to improve the effect of the treatment of acute myocardial infarction(AMI).Methods 1 Separated and cultured BMSCs by the whole bone marrow culture,and 5-azacytidine induce into cardiomyocyte cells. Before and after of induction, identified the cells by the expression of cells surface marker CD44 and CD34; and c Tn T by immunocytochemistry. 2 Left anterior descending coronary arteries were ligated to produce AMI model, and forty Sprague-Dawley rats at the beginning of weaning were randomly divided into four groups: 1) control group, one week before the modeling,every rat is gave 2ml normal saline intragastric administration,1 time/day,1 week;after the modeling,at the infarct area and around,divides five points injecting DMEM culture medium.2) drug group: one week before the modeling,every rat is gave 2ml Rosuvastatin solution(1mg/ml) intragastric administration,1 time/day,1 week;after the modeling,at the infarct area and around, divides five points injecting DMEM culture medium.3)transplantation group: one week before the modeling,every rat is gave 2ml normal saline intragastric administration,1 time/day,1 week;after the modeling,at the infarct area and around,divides five points injecting 2×106 BMSCs.4)combining group: one week before the modeling, every rat is gave 2ml Rosuvastatin solution(1mg/ml) intragastric administration,1 time/day,1 week;after the modeling,at the infarct area and around,divides five points injecting 2×106 BMSCs.3 After the modeling, observing the changes in electrocardiogram and the picture of myocardium H.E to determine the success of AMI models.To monitor the expression of IL-8 、 TNF-α and cardiac troponin T(c Tn T) after operate 8 hours, the method is to extract blood from aorta abdominalis and separate serum.To weigh the whole heart and ventriculus sinister and calculate the left ventricular hypertrophy(LVH) after operate 4 weeks. Immumofluorescent staining is to detection the survival rate of the transplanted SMSCs. All date were analyzed by the SPSS17.0. The measurement data were demonstrated by(mean ± SD). The one-way ANOVA is used in two measurement data compared. T tests was used in two measurement data. A value of P<0.05 was considered as statistical significant.Results 1 The cell morphology of primary cultured cells were showed in fusiform, or irregular shape, eventually were fusiform, arranged in a direction. After passage, BMSCs growth evenly distributed, more single morphology, and growth speeder. The immunohistochemical showing, the positive expression of CD44, the negative expression of CD34. 2 Ligating left anterior descending artery of rats, the myocardial of ligation around and below turn to tan-white immediately, the change of ECG ST-elevation is raised arch upwaid, and returns to baseline slowly.3 the expression of IL-8、TNF-α and c Tn T:compared with control group, drug group, transplantation group and combining group are decreased of IL-8、TNF-α and c Tn T; Compared with drug group and transplantation group, the combining group is decreased more obviously; Differences are statistically significant(P<0.05).4 the result of left ventricular mass and the percent to whole heart:drug group, transplantation group and combining group are lower than control group, the combining group reduced more significantly than drug group and transplantation group;Differences are statistically significant(P < 0.05).5 the result of immunofluorescent staining: Compared with transplantation group and combining group, combining group’s cells of the nucleus is blue-fluorescence are more than transplantation group’s.Conclusions 1 Through the induce of 5-azacytidine, BMSCs can be obtained to cardiomyocyte cells. 2 Rosuvastatin through anti-inflammatory 、 antioxidant stress to improve the survival rate of the transplanted SMSCs. 3 Rosuvastatin combines with transplantation of BMSCs can improve the effect of the treatment of AMI.
