Objective: Explore the affection to acute myocardium infarction myocardial tissuetoll-like receptor4(TLR4) protein expression by transplantation of autologous bonemarrow stromal cells through vein.Methods:60healthy New Zealand rabbits were randomly divided into control group,surgery group and treatment group, each group of20. The surgery group and treatmentgroup are carried out ligation of thoracic and established acute myocardial infarctionmodel at1/2middle of left anterior descending coronary artery, and the control group iscarried out thoracotomy in the same place and with the penetration of surgical sutures,but not ligatured.2weeks after the membrane, the treatment group is implantedautologous bone marrow stromal stem cells through the ear marginal vein, and surgerygroup and control group are implanted slow-release phosphate liquid with the sameamount as bone marrow stromal stem cells;4weeks after transplantation, the hearts ofeach group are taken out respectively, the TLR4expression in myocardial tissue of threegroups is tested by Western blotting detection, and calculate the relative quantitative ofTLR4through the Gel-Pro Analyzer software testing protein bands grey value.Comparison between groups adopts Kruskal-Wallis Test, and data is expressed usingmedian and the percentile (upper quartile, lower quartile), the difference with P <0.05has statistical significance.Results:The TLR4expression within the Ischemic necrosis of myocardial tissue of thetreatment group is obviously higher than that of the control group (P <0.05), The TLR4expression within the infarcted myocardial of the treatment group is lower than surgicalgroup (P <0.05), but higher than control group (P <0.05).Conclusion:1.TLR4expression rapid rise after the occurrence of acute myocardialinfarction prompts that TLR4is of great importance in the in the onset of acutemyocardial infarction, it may be as important inflammatory injury of ischemia of inflammatory receptor mediated myocardial tissue;2. Autologous bone marrow stromalstem cells can survive and differentiate after transplantation in acute myocardialinfarction tissue;3. The transplantation of autologous bone marrow stromal stem cellsvia ear marginal vein can treat acute myocardial infarction, one of its mechanisms maybe related to inflammatory reaction induced by inhibiting TLR4, it can partly inhibit thesynthesis and release of inflammatory factors of the inflammatory reaction induced byinhibiting TLR4.
