Our experiment planed to add a new type of pH dependent material(p H-dependent material, pHDM) into the PLGA. This material was prepared by the resin with the method of induced synthesis. And the material could degrade in high-speed within a specific pH range of 6.6-6.8. It had not only good mechanical strength but also fast degradation rate. As could make contained drugs released rapidly to promote tissue regeneration and proliferation when tissue anoxic happened. The threedimensional pH-DM/PLGA scaffold was prepared with the two kinds of materials by electrostatic spinning.The results showed that: 1. the best conditions to prepare the pH-DM/PLGA scaffold was as follow: quality ratio of PMMA to EBA is 9:1, reaction temperature was 60 °C, and BPO concentration was 10 %. 2. The optimal process to prepare pH-DM/PLGA was as follow: material concentration was 5%, injection rate was 0.4 mL/h, receiving distance was 14 cm, and voltage is 11 kV. The fiber in the scaffold had similar diameter, the porosity was 79.03 %, the mechanical properties were as follows: The tensile strength was 760.8±26.8 KPa, tensile rate was 0.393±0.015, bursting strength was 764.6±24.9 KPa. The pH sensitivity of pH-DM was still remained after high voltage electrostatic spinning. 3. The hemolysis test results showed that the hemolysis rate of the scaffold material was less than 5%. 4. Mice abdominal cavity implantation experiments showed that, the tissue cells grew along the nanometer fiber’s configuration. Some tissue differentiated into new blood vessels. And the nanofibers were most degraded in 30 days.We drew the conclusion that the prepared pH-DM/PLGA scaffold had suitable blood compatibility histocompatibility and biodegradability. It was a good artificial blood vessel scaffold.
