Pi-deficiency is common in Chinese medicine syndromes of the digestive system based multi-system of multiple organ dysfunctions.Pi-yin-deficiency which is one of sydroms of Pi-deficiency is caused by internal injuries of qi and blood,thus leading to the damage of body balance between the various organs and leading to dysbacteriosis.In this research,intestinal flora DNA which was extracted directly from rat feces was amplifed using primers of ERIC and 16S rDNA to generate ERIC-PCR and 16S rDNA-RFLP genomic fingerprints."Pi-yin-deficiency" model rats were induced by water extract of Folium sennae & Thyroxine.By comparing the ERIC-PCR and 16S rDNA-RFLP fingerprints during healthy and diseased phases, the molecular evaluating system was established.Four "Pi-deficiency" therapy Chinese herbal recipes:Si-jun-zi-tang,Li-zhong-tang,Bu-zhong-yi-qi-tang and yi-wei-tang were evaluated using the molecular evaluating system.The results showed that Si-jun-zi-tang,Li-zhong-tang,Bu-zhong-yi-qi-tang had better effect than yi-wei-tang.Three molecular narkers were cloned to further clarify Pi-yin-deficiency related bacteria whose physiological role were discussed either.The gene fingerprint can reflect the overall changes of intestinal flora.This study reflects the advantage of combining of modern biotechnology and TCM.
