Objective:Analysis the intestinal flora diversity of the normal hilits in traditional Uighur medicine comparison of species differences in intestinal micro flora of four groups. According to the Uighur medicine,The Healthy subjects were divided into four hilits then We used ERIC primers to generate community fingerprints and Denaturing Gradient Gel Electrophoresis(DGGE) to analyze the different intestinal microbial and obtain intestinal microflora unique fingerprint of four groups, comparing groups intestinal microflora distribution and differences. Methods:①According to the Uighur medicine,206 Healthy subjects were divided into Khan Hilit(53), Balgham hilit(51), sapra hilit(50), sawda hilit(52) four groups,Fecal samples were collected from these healthy individuals,The total genomic DNA was extracted.The total DNA were amplified by using the ERIC primers to generate community fingerprints.Anlaysis each groups Shannon-Weiner index and Jaccad index, get four normal humor qualitative person intestinal flora distribution of similarity and diversity.②The total genomic DNA was extracted from each sample and used for PCR amplification of V6-V8 regions of 16S rDNA. Subsequently,DGGE analysis of the amplified sequences was performed. Then we compared the intestinal floras similarity and the diversity of all subjects. In order to elucidate the bacteria represented by DGGE bands, we cloned the DGGE bands and selected randomly positive clones for sequencing. Results:①The number of DNA bands of Khan Hilits more than the other three groups (P< 0.05), The number of DNA bands of Balgham hilit less than other three groups (P< 0.05). The diversity of intestinal microbial communities of Khan Hilits has been increased(P< 0.05) and the intestinal flora of Balgham hilit has been reduced(P< 0.05) compared with another groups in The ERIC-PCR②The number of DNA bands of Khan Hilits more than the other three groups (P< 0.05), The number of DNA bands of Balgham hilit less than other three groups (P<0.05).③The sequences that frequency much can representative each sample were compared with sequences in the Genebank nucleotide sequence database by using Blast software and phylogenetic trees were constructed.Conclusion:①There were some differences in intestinal microbial communities of different normal hilit groups.②Initialy construct phylogenetic trees of intestinal flora diversity of the normal hilits in traditional Uighur medicine。...
