Pi-deficiency, a clinical syndrome in traditional Chinese medicine, is one of the most common digestive system diseases. We attempted to introduce ERIC-PCR and 16S rDNA-RFLP to analyze the variety of rat intestinal microflora, and helped to build a better understanding of Pi-deficiencyIn this research, total intestinal microflora DNA which was extracted directly from rat feces was amplified using primers of ERIC and 16S rDNA to generate ERIC-PCR and 16S rDNA-RFLP genomic fingerprints. The pharmacodynamic evaluating system, including identified molecular markers (net integral area and abundance of DNA band), Shannon’s index for diversity of intestinal microflora, and Sorenson’s pairwise similarity coefficient, was established. With the established molecular evaluating system, we compared the therapeutic effects of four clinical commonly used Pi-deficiency therapy TCM (Si-jun-zi-tang, Li-zhong-tang, Bu-zhong-yi-qi-tang and Yi-wei-tang).We have used molecular biology and genomics method to investigate Pi-deficiency and its treatment, and found the molecular evaluating system in this research. It’s also provided a new way of thinking and the method to further explore Pi-deficiency syndrome and TCM evaluating and screening.
