Parkinson’s disease is a common central nervous system disease in older people. Rigidity, tremor, bradykinesia and abnormalities in body posture and skilled movements is the main clinical symptom. Ginkgo biloba leaves has therapeutic effects, however preparation which is made of the major components of Ginkgo biloba for treatment of Parkinson’s disease has not been reported.In this study, using HPLC determin the content of quercetin in Ginkgo biloba, using orthogonal design to choice the optimum of extracting the flavonoids in Ginkgo biloba by Ethanol. Solvent quantity and temperature have significantly impact.13 times the amount of 70% ethanol,80℃, extract 1 hour and extract two times is optimum.Using centrifugal, macroreticular resin chromatography, the second extraction to purify of the extract, The total flavonoids in the purity of the Ginkgo biloba extract is greater than 16.8% and bilobalide is greater than 2.1%. And macroreticular resin purification process was optimized. Stock solution was adjusted pH to 3, washed with water,4BV 70% ethanol elution. Second extraction with anhydrous ethanol-ethyl acetate (3:7), solid-liquid ratio of 1:20 is the best.The quality of the extracts was studied using the TLC. Extract and ginkgo leaf under 365nm, the corresponding position of main spots have the same color of fluorescence. Determining the flavonoids content of extact by UV-Vis spectrophotometry. Determining the bilobalide content of extract by TLCS.By measuring the different prescriptions of Ginkgo biloba extract gel’s gelation temperature, viscosity and in vitro release conditions, choice the best prescription gel. With mass fraction of 18% poloxamer 407,4% poloxamer 188,10% Ginkgo biloba extract,2% HPMC is the best, its gelation temperature over 30℃, in vitro release of nearly constant speed.Established rat model of Parkinson’s disease using the 6-OHDA, slicing of brain and immunohistochemistry result showed that ginkgo biloba extract fed group and the Ginkgo biloba extract gel intranasal administration group of Parkinson’s model rats have therapeutic effects.
