This paper reported the water soluble phytochemical components investigation on Dendrobium candidum. Coupling with chromatography techniques and spectrum methods, this paper extracted and identified eight coumpouds on Dendrobium candidum. Including two lignans, syringaresinol-4,4’-O-bis-β-D-glucoside (1) and (-) syringaresinol (2), three phenylpropanoids glycosides, (1’R)-1’-(4-hydroxy-3,5-dimethoxylphenyl) propan-1’-ol-4-O-β-D-glucopyranoside (5) and shashenoside I (6), dendrocandinoside (7) and three phenolic glycosides, khaephuoside A (3), 1-mether-2-O-[β-D-apiofuromosyl-(1â†'2)-β-D-glucopyranside] (4) and leonuriside A (8). Among them, compound 4 and 7 are new compounds. Compound 2 was isolated from Dendrobium candidum for the first time, and others were isolated from the Dendrobium species for the first time.Also, in this paper, a new method for simultaneous determination of eight major components on Dendrobium candidum, including one lignan glycoside, (+)-syringaresinol-β-D-glucopyranoside, one amide, moupinamide, one flavanone, naringenin, five phenols,4,4’-dihydroxy-3,5-dimethoxybibenzyl, dendrophenol,3,4’-dihydroxy-5-methoxybibenzyl, gigantol and 3,4-dihydroxy-5,4’-dimethoxybibenzyl, using high-performance liquid chromatography (HPLC)-diode array detection (DAD) was described. The separation of the eight compounds was effected by RP-HPLC (Phenomenex Kinetex C18 column,100 mm×4.6 mm i.d.,2.6μm) using linear gradient elution systems of acetonitrile-5% acetonitrile including 0.1% trifluoroacetic acid (TFA). Satisfactory separation of these compounds was obtained in less than 35 min. The method was validated for linearity, repeatability, limits of detection (LOD) and limits of quantification (LOQ). Good results were obtained with respect to repeatability (RSD< 2.4%) and recovery (95%-105%). The developed method was applied to the analysis of 8 major components from totally 90 Dendrobium species, and referred a standard to the quality control of Dendrobium species.
