Propolis is a mixture of colloidal solids with aromatic fragrance, consists of gum which bee collected from plant spores and tree’s trunks, bees wax and other maxillary gland secretions.A group of bees only product 0.2 to 1.2g propolis one day,named as "purple gold" by scientists, is very precious. A lot of research and clinical practice showed that propolis is rich in bio-active substances,so is widely used to improve immunity, reduce blood sugar, anticancer and other features of the development of health foods in recent years.Currently, propolis extraction method mainly was ethanol extract, however, the propolis’s ethanol extracts (EEP) was black or reddish brown, and smell bad, difficult entry, even lead to allergies,high content of heavy metal of lead. With the continuous development of extraction methods,supercritical CO2 extraction of propolis came into being, it’s extracts is pale yellow, pure taste,low lead content, to avoid some traditional defects from the ethanol extraction process and was closely watched.Propolis resources as valuable and rich in bio-active substances, therefore, establish a more convenient method to identify the quality of SEP and further development of the biological activity of SEP are more significance to constantly open up the market for propolis and monitoring the quality of propolis in the market.Thus,we set up the fingerprints of supercritical CO2 extract of propolis using GC which collected from different regions of China, then determined total flavonoids in SEP and EEP using spectrophotometer, and finally we studied the effect of SEP to two kinds of experimental gastric ulcer. The results show that:(1)The chromatograms obtained from the 16 SEPs include nine common peaks,their relative retention times are 6.831s,8.447s,10.662s, 13.524s,17.678s,22.94s,30.094s,52.62s and 58.158s,their average peak area are 237.930,998.088,14068.630,1699.602,37875.459,1108.638, 20148.995,9783.739 and 3251.770 and the ratio of average peak area are 6.2,1.0,14.1,1.7,37.9,1.1,20.2,9.8 and 3.3 and their atlas information make up of the SEP fingerprints.(2) Total flavonoids of Supercritical CO2 extract of propolis (SEP) are between 0.4 to 1.1%.The highest totol flavonoids of SEP was Yunnan province’s propolis, and the content accounted for 1.1% of propolis; Hunan and Hubei province had the middle content of flavonoids in SEP; Heilongjiang region generally had low content of total flavonoids in SEP, only accounted for 0.4% in propolis;Henan and Sichuan province in the flavonoid content in SEP had a relatively large gap;the total flavonoids from SEP were lower than EEP.(3)Through animal tests we found that the resist proportion of water stress of gastric ulcer were respectively 66.11%,78.24% and 74.48% to SEP(30mg/kg,10mg/kg and 3 mg/kg) and 64.43%,to EEP(10 mg/kg); the resist proportion of ethanol-induced gastric ulcer were 69.37%, 55.86% and 78.83% to SEP(30 mg/kg,10 mg/kg and 3 mg/kg) and 66.86% to EEP(10 mg/kg) respectively.(4)The blood MDA content in ethanol-induced gastric ulcer was significantly (P<0.05) reduced from 5.11 nmol/ml to 2.66nmol/ml by treatment with SEP (3mg/kg);the blood SOD activity in ethanol-induced gastric ulcer were significantly (P<0.05) increased from 161.78 U/ml to 238.16U/ml,278.09U/ml and 239.32U/ml by treatment with SEP(10 mg/ kg and 3 mg/kg) and EEP(10 mg/kg) respectively.
