Objective To investigate an association between heat stress protein 70(HSP70) and Tumor necrosis factor-α(TNF-α) gene polymorphism and the psychological stress among Hui and Han nationalities in Ningxia; To explore the genetic susceptibility to chronic psychological stress, clarify the genetic basis of chronic psychological stress and susceptibility genes.Method According to a case-control study design, workers of regional organizations and institutions aged 20-60 who received medical health checkup in General Hospital of Ningxia Medical College from January 2012 to October 2013 were enrolled in the study. According to the inclusion and exclusion criteria, 622 patients were identified as a case group; population from the same period in General Hospital of Ningxia Medical College out-patient clinics for regular health check-randomly selected 600 healthy people in the control group. Data were collected through questionnaire, physical examination and laboratory examination, and Minor Groove Binder-Taqman(MGB-Taqman) probe method was applied to detect the single nucleotide polymorphisms of HSP70-1-110, +190 and HSP70-2 +1267, +2074 and HSP70-Hom +2437 and TNF-α-308.Results Differences of mild, moderate and severe stress level between the group of three groups of WC, WHR, SBP, DBP, FPG, TG, HDL- C, AST, UA were statistically significant(F = 5.888, 9.705, 11.688, 9.705, 13.955, 5.288, 7.554, 4.612, 7.017, P < 0.05 or P < 0.01), with the increase of stress level, WC, WHR, SBP, DBP and UA are in decline after rising first, the TG showed a trend of decline after rising first, FPG, AST are on the rise trend,HDL-C a downward trend, age, BMI, TC, LDL-C, ALT in three groups had no significant difference between the stress level groups(all P > 0.05);Cases of different stress levels in the group of HSP70-1 gene-110 between genotype frequency difference was statistically significant(P<0.05);The control group in different stress levels between groups of HSP70-1 gene- 110 genotype frequency difference was statistically significant(P<0.05);Control group in different stress level of HSP70-hom gene + 2437 locus allele frequency difference was statistically significant(P < 0.05);The control group in different stress levels of TNF-α gene-308 loci genotype and allele frequency difference had statistical significance(P < 0.05);Different stress level set of the general population HSP70 + 190-1 gene loci genotype frequency difference was statistically significant(P < 0.05);Different stress level set of the general population HSP70 + 2074-2 gene loci genotype and allele frequency difference had statistical significance(P<0.05);HSP70 in cases of group HSP70-1 gene + 190 loci, HSP70-1-110 loci and HSP70-2 + 1267 gene loci of three level of psychological stress difference between genotype groups were statistically significant(all P <0.05).Conclusion HSP70-1 gene –110 loci and +190 polymorphic loci associated with ningxia crowd psychological stress susceptibility might exist;HSP70-2 gene + 2074 loci and TNF-α-308 loci genotype and allele distribution differences between different stress level group, prompt the correlation between gene polymorphism and stress;HSP70-hom gene+2437 loci alleles may be associated with chronic psychological stress; in cases of group HSP70-1 gene + 190 loci, HSP70-1-110 loci and HSP70-2 +1267 gene loci of the three psychological stress level differences between genotype groups, prompt HSP70 gene may be vulnerable to stress.
