| Objective: This thesis, on one hand is to discuss whether a six weeks`intervention of swimmingand metformin can make a combined synergistic effect in improving LKB1-AMPK-GLUT4signal pathway in the rats of type2diabetes that induced by the combination fo a high-fat dietand a low-dose STZ, and on the other hand is to provide scientific theoretical basis for clinicaltreatment of type2diabetes.Methods: Fifty-five SD rats was randomly divided into five groups, it were normal controlgroup(NC)ã€diabetes control group(DC)ã€diabetes exercise group(DE)ã€diabetes medicinegroup(DM)ã€diabetes exercise and medicine group(DEM); The NC was feed with normal dietand all the diabetic groups were feed with high-fat diet; A low-dose STZ was given to all groupsof diabetes to build the model of type2diabetes after seven weeks, and then the DE group wasgiven six week`s swimming intervention, the DM group was given six week`s metforminintervention, the DEM group was given a six week`s combined intervention of swimming andmetformin. After six weeks, the levels of blood glucoseã€insulinã€Kittã€TGã€FFAã€MGã€P-LKB1ã€P-AMPKa12ã€AMPKa2ã€P-AMPKa2ã€GLUT4in all groups should be tested.Results: Compared with NC, the levels of blood glucoseã€insulinã€TGã€FFA are significantlyrising in the DC(P<0.01ã€P<0.01ã€P<0.01ã€P<0.01), the levels of Kittã€MGã€P-LKB1ã€P-AMPKa12ã€AMPKa2ã€P-AMPKa2ã€GLUT4are significantly descending in DC(P<0.01ã€P<0.01ã€P<0.05ã€P<0.05ã€P<0.05ã€P<0.01ã€P<0.01); Compared with DC, the levels of insulinã€TGã€FFA are significantly descending in DE(P<0.05ã€P<0.05ã€P<0.05), the level ofP-AMPKa2ã€GLUT4is significantly rising in DE(P<0.05ã€P<0.05),the levels of bloodglucoseã€insulinã€TGã€FFA is significantly descending in DM(P<0.05ã€P<0.05ã€P<0.05ã€P<0.05), the levels of Kittã€MG are significantly rising in DM(P<0.05ã€P<0.05), the levelsof blood glucoseã€insulinã€TGã€FFA are significantly descending in DEM(P<0.01ã€P<0.01ã€P<0.01ã€P<0.01), the levels of Kittã€MGã€P-LKB1ã€P-AMPKa12ã€AMPKa2ã€P-AMPKa2ã€GLUT4are significantly rising in DEM (P<0.01ã€P<0.01ã€P<0.05ã€P<0.05ã€P<0.05ã€P<0.01ã€P<0.05); Compared with DE, the levels of blood glucose is significantly descending in DEM(P<0.05).Conclusions: The intervention of exercise〠metformin and combination of exercise andmerformin can make a different degrees impact for in improving glucose and lipid metabolism disorder of type2diabetes, the combined intervention of exercise and metformin can effectivelyrepair the injury of LKB1-AMPK-GLUT4signal pathway in the skeletal muscle of type2diabetes, and the intervention of simple exercise can partly repair the injury of AMPK-GLUT4signal pathway, and the intervention of simple metformin make no difference for the injury ofLKB1-AMPK-GLUT4signal pathway. |
