Objective: The purpose of this paper is to through the observation of aerobic exercise on age of rat skeletal muscle ROS, IL- 6, the nf-kappa B and AMPK expression level change and the influence of IL- 6 expression levels in the blood, the explanation of the inflammatory response in the skeletal muscle model, explore about the ROS and the possible mechanism of IL- 6 in AMPK pathway, provide theoretical basis for the research of anti-agingMethods: 48 male SD rats were randomly divided into: youth group, youth group, control group of middle-aged and young, middle-aged and young sports group, middle-aged group, middle-aged group, each group of eight. Experiment using improved aerobic exercise model: according to Bedford aerobic exercise model, the reference load intensity(treadmill grade of 0 °, velocity of 19 m/min, equivalent to 70% VO2max), training for 6 days a week, once a day, 60 minutes at a time, for aerobic training for 8 weeks, 8 weeks after the training materials. By ELISA to detect serum of rats and the content of the IL- 6 in skeletal muscle tissue; DHE fluorescent staining method are used to observe ROS expression in rat skeletal muscle tissue level; Western blotting technology to detect the nf-kappa B in rat skeletal muscle tissue and differences in the levels of AMPK proteins expression; Real time quantitative PCR technique to detect IL- 6 in rat skeletal muscle tissue, the nf-kappa B and AMPK gene mRNA expression level.Results:Increasing age in rat skeletal muscle and serum, the content of IL- 6. Aerobic exercise training within eight weeks after exercise group in rat skeletal muscle and serum IL- 6 levels higher than the control group. Aerobic exercise training within eight weeks after exercise group rats skeletal muscle ROS levels lower than the control group. Increasing age increased the nf-kappa B expression in rat skeletal muscle. Aerobic exercise training within eight weeks after rat skeletal muscle nf-kappa B expression is lower than the control group. Increasing age P- AMPK and AMPK expression in rat skeletal muscle. Aerobic exercise training within eight weeks after exercise group P- AMPK and AMPK expression in rat skeletal muscle were higher than the control group.Conclusions:Through aerobic training can reduce rat skeletal muscle ROS expression,so as to reduce skeletal muscle oxidation damage. The bones in the process of aging muscle mitochondrial respiratory function decrease, increase the production of ROS, so that the body into the high oxidative stress status,increased expression of IL-6. Aerobic exercise training intervention, can improve the function of skeletal muscle mitochondria, decreasing the production of ROS, reduce the oxidative stress state of organism.Aerobic exercise can play to reduce expression of the aging process of aging rats NF-κB, help to slow the aging skeletal muscle atrophy.Aerobic exercise in aged rats, can through the regulation of nuclear NF- κ B expression and increasing AMPK expression, so as to achieve the regulation of stress state, play a role in delaying aging, the specific regulation study needs to be further improved.
