Objective: To research the molecular mechanism of hepatic lipid metabolism in obesity,explore the molecular mechanism of exercise,diet and exercise combined with diet three kinds of intervention methods on the adiponectin-APPL1-LKB1-AMPK signaling pathway regulates lipid metabolism,and provide a theoretical basis for the prevention and treatment of obesity caused by exploring methods of lipid metabolism disorders of the liver.Methods: 54 C57BL/6J mice(3 weeks old)were randomly divided into normal group(n=17)and high-fat group(n=37)and given ordinary feed and high-fat feed respectively.After 12 weeks of feeding,the normal group was divided into NC(n=8)and NE(n=9),and the successfully modeled 34 mice in the high-fat group was divided into HC(n=8),HE(exercise intervention,n=9),HN(diet intervention,n=8)and HNE(exercise and diet intervention,n=9)according to their weight.Diet intervention means changing the ordinary feed into high-fat feed.In exercise intervention group,after two weeks of adaptive treadmill training,the training speed is fixed to be 20m/min for 60 min,6 days a week from the third week,and all interventions lasted for 8 weeks.When the interventions were over,serum Adiponectin by ELSA,colorimetry liver TG and FFA,Real-Time PCR and Western-Blot were messured in Adiponectin-APPL1-LKB1-AMPK signaling pathway in liver tissue to determine the relevant indicators of gene transcription and protein expression levels.Results:1.Compared with NC,HC group serum adiponectin decreased significantly(p <0.01),FFA and TG were significantly increased(p <0.01).Compared with HC,NH and HE serum adiponectin were significantly increased(p <0.01),liver FFA,TG decreased(p <0.01 or p <0.05).Compared with HN,HNE serum adiponectin increased(p <0.05),FFA and TG decreased(p <0.01 or p <0.05).Compared with HN and HE,HNE levels of serum adiponectin increased(p <0.05),FFA and TG decreased(p <0.01 or p <0.05).2.Compared with NC,the levels of adiponectin,APPL1,LKB1,AMPK protein and m RNA in HC were decreased(p <0.01),ACC level was increased(p <0.05).The levels of adiponectin,LKB1,AMPK and APPL1 in HN,HE were significantly higher than those in HC(p <0.05 or p<0.01).The group which was on the diet and exercise adiponectin,APPL1,LKB1,AMPK the levels of protein and m RNA were higer than HN(p <0.01 or p <0.05),but the ACC was completely the opposite(p <0.05).Compared with HE,the expression of adiponectin,AMPK and ACC protein in HNE was significantly higher than that in HE(p <0.05 or p <0.01),while APPL1 and LKB1 were decreased(p <0.01 or p <0.01),in terms of m RNA the adiponectin,APPL1,LKB1,AMPK and ACC of HNE decreased(p <0.05 or p <0.01).Conclusion:1.One of the prime reasons of liver lipid metabolism is the long-term high-fat diet,which reduces serum and liver Adipo levels,inhibiting the Adiponectin-APPL1-LKB1-AMPK signaling pathway-related protein and m RNA expression.2.Abnormal lipids metabolism caused by obesity can be improved by long-term moderate aerobic exercise,diet and exercise combined with dietary,which can up regulate levels of Adipo in the liver activate the Adiponectin-APPL1-LKB1-AMPK signaling pathway protein and m RNA expression.
