Experimental Objective:1. To explore the effect of treating non-alcoholic fatty liver disease by activating AMPK-ACC-MA pathway using motion.2. To study whether the traditional Chinese medicine used in the experiment can activate AMPK-ACC-MA pathway.3. To observe if the two together has any effect on NAFLD rats’fat metabolism.4. To provide new ideas and treatments for the treatment of NAFLD.Methods:605-week-old male SD rats were used in the experiment. Those rats were divided into quiet control group (C group), high-fat diet group (HF group), high-fat diet with traditional Chinese medicine interfering group (HZ group), high-fat diet with exercise interfering group (HE group), and high-fat diet with exercise and traditional Chinese medicine interfering group (HZE group). The rats in HE group and HZE group do a60-minute swimming without load every afternoon. They rest on Sundays. The whole process continues for10weeks. The rats in HZ and HZE group take in1ml/100gBW doses of traditional Chinese medicine each morning and all other groups take in the same amount of saline instead. After10weeks, the content of serum TG, TC, AST, ALT, FFA and HDL were detected. The activity level of liver ACC and MA were also detected. The expression of liver AMPKal and AMPKa2mRNA were examined using real-time quantitative PCR. The histological changes of livers were observed under light microscope after HE staining.Results:1. After HE staining the rat’s liver, it shows under the light microscope that the rat livers in C group do not have lipid droplets vacuoles. The central vein is radially arranged. The nucleus is located in the central cell. There is almost no inflammatory cell infiltration. The sinusoidal is visible and clear. The rats’ liver cells in HF group have more lipid droplets vacuoles. The cells swell and accompanied by inflammatory cell infiltration. The sinusoidal is narrow. The nucleus is pushed to one side. The cells in HE group have less lipid droplets vacuoles. The nucleus is at the center of the cell. The central venous slightly swell. The cells in HZ group have more lipoid droplet vacuoles than HE group. Most of the nucleus is at the center of the cells. The sinusoidal narrows and the gap are small. The gap among the sinus is normal for cells in HZE group. There are a small amount of lipid droplets vacuoles.2. Compared with C group, rats in HF group have much higher liver weight, body weight, and liver and body weight ratio (P<0.01). Aerobic exercise and traditional Chinese medicine can significantly control the growth rate of liver weight and the liver and body weight ratio (P<0.01). The effect of interacting aerobic exercise and traditional Chinese medicine has a great influence on rats’liver wet weight and body weight (P<0.01). The effects on liver and the ratio of body weight are significantly different (P<0.05).3. The content of serum TC and FFA in HF group is significantly higher compare with C group (P<0.01). There is a significant difference of the content of serum TG (P<0.05). Traditional Chinese medicine can influence the content of serum TC in rats significantly. Supplement traditional Chinese medicine can effectively reduce the content of serum FFA (P<0.05). Aerobic exercise, traditional Chinese medicine and the effect of interacting aerobic exercise and traditional Chinese medicine can further reduce the content of serum FFA (P<0.01). When it comes to the indicator of serum TG in rats, traditional Chinese medicine can effectively reduce the content of serum TG (P<0.01). Also, aerobic exercise, traditional Chinese medicine and the effect of interacting the two can also reduce the content of serum TG (P<0.05).4. The activity level of rats’ serum AST and ALT in C group are significantly lower than HF group (P<0.01). There is a significant difference in serum AST activity level between HF group and C group (P<0.05). Aerobic exercise and traditional Chinese medicine have a great influence on reducing the content of serum ALT (P<0.01). Aerobic exercise together with traditional Chinese medicine can effectively reduce the content of serum ALT (P<0.05). Supplement traditional Chinese medicine can significantly reduce the content of serum AST (P<0.05). Aerobic exercise can effectively increase the content of serum HDL and the effect if significant.(P<0.05).5. Compare to the quiet control group, the content of ACC in liver is significantly different between the quiet control group and the high-fat diet group (P<0.05). The content of MA in C group is much lower than it in HF group (P<0.05). Analysis found that traditional Chinese medicine can effectively reduce the content of ACC and MA in rats’ livers (P<0.01). Aerobic exercise can decrease the content of ACC and MA in rats’ livers (P<0.05). Traditional Chinese medicine and aerobic exercise together can influence the content of MA in livers (P<0.05).6. AMPKalmRNA expression in each group has no significant difference. Comparing to C group, the expression of AMPKalmRNA in HF group is significantly low and has a significant difference (P<0.01). Aerobic exercise, traditional Chinese medicine, aerobics exercise interact with traditional Chinese medicine all have no effective influence on the expression of liver tissue AMPKalmRNA. The expression of the liver tissue AMPKa2mRNA is influenced by aerobic exercise and aerobic exercise and traditional Chinese medicine’s interaction effects. The effect is significant (P<0.05).Conclusion:1.10weeks of feeding non-alcoholic fatty liver rats’ replication model with high fat diet success.2. Aerobic exercise and traditional Chinese medicine have significantly reduced the level of fatty liver disease in high-fat diet rats. At the same time, joint intervention of these two has a significant effect.3. Aerobics and traditional Chinese medicine can effectively improve fat variable degree in rats with non-alcoholic fatty liver by regulating lipid metabolism.4. Aerobic exercise can significantly improve the expression of rat liver tissue AMPKa2mRNA to modulate the liver lipid metabolism. Aerobic exercise and traditional Chinese medicine can significantly elevate the expression of AMPKa2mRNA in rat liver tissue.
