The purpose of the experiment:To investigate the effect of high fat diet on testicular germ cell apoptosis in male rats, and reveal the intervention of aerobic exercise on testicular germ cells apoptosis in male rates on high fat diet.The Method of the experiment:32SD male rats were randomly divided into4grops:control group (C),high fat diet group(HF),the common movement(CE),high fat diet group (HE),each group has eight rats; Group C and CE group was given common feed, HF group and HE group were fed with high fat, high heat. Kinematic scheme for10weeks60min without weight loading swimming movement, six days a week. After the end of the experiment detect serum TC, TG, HDL-C, LDL-C, MDA, SOD, using the ELISA method determination of T, FSH, LH, Caspase-3, Cytc, using flow cytometry to detect sperm apoptosis of rats, real-time fluorescent quantitative detection Caspase-3mRNA in rat testicular tissue and Cytc mRNA expression, then observe morphological changes of testicular tissue.The result of the experiment:1. After observe rat testis tissue morphology through HE staining under light microscope normally and orderly developed seminiferous tubules spermatogenic cells can be found. Seminiferous tubule in CE group of gap widens and mature sperm can be found in the tube. Testicular tissue seminiferous tubule wall became thinner, spermatogenic cells decrease in the number and in disordered arrangement, The mature sperm number decreased, occasional cell shedding phenomenon can be seen, individual rats Spermatogenic cells and detached from basement membrane in some cases, fall off sperm cells is formed in the lumen. HE group gave birth to the small tube born normal sperm cells basic development at all levels and order, a large number of mature sperm can be seen, cell falls off phenomenon, duality can be born seminiferous tubule interstitial tissue is loose.2. CE group compared with group C, the body weight significantly decreased, fat significantly reduced testosterone week, TG significantly reduce, HDL significantly increased, Lee’s index, the total number of sperm, deformity rate and activity rate, TC, and LDL there was no significant difference. HF group compared with group C, fat weight, Lee’s index, testosterone weeks and malformation rate significantly increased, the total number of sperm significantly reduce, activity rate significantly decreased, TC, LDL significantly increase, TG、HDL, no obvious difference. HE group compared with HF group, weight fat significantly lower testosterone week, Lee’s index decreased significantly, deformity rate significantly decreased, activity ratio increases, the TC, TG, HDL, LDL and there were no significant difference on the total number of sperm.3.CE group compared with C, a significant rise in rat serum level of T, LH and FSH levels significantly increased. HF group compared with group C, the rat serum T, LH, FSH were significantly reduced. HE group compared with HF group, the rats serum T level was increased, but the difference was not significant. The marked increase in LH and FSH level. Through the analysis of variance, the high fat diet can cause rat serum LH and FSH levels, elevated serum LH and FSH levels as a result of aerobic exercise.4. CE group compared with group C, the activity of SOD in serum of rats significantly increased, MDA content decreased significantly; HF group rats serum SOD activity significantly reduce, the content of MDA significantly increased, HE than HF group serum SOD activity significantly increased, MDA content decreased significantly. Through the single factor analysis of variance, the high fat diet can lead to cut down the content of SOD and MDA in serum of rats content increased, while aerobic exercise can increase SOD content and decrease MDA content.5.Apoptotic cells in rat testicular tissue:compared with group C, CE group sperm apoptosis significantly reduced testicular tissue and HF sperm apoptosis significantly increased in group of testicular tissue. HE group compared with HF group, the sperm apoptosis decreased significantly in testicular tissue. Through the analysis of variance, the high fat diet can increase sperm apoptosis in testicular tissue, and aerobic exercise can ease the situation.6.CE group compared with group C, Caspase-3in testicular tissue of rats and Cytc are lower but no differences, Caspase-3and Cytc mRNA expression quantity significantly reduced. HF group compared with group C, Caspase-3in testicular tissue of rats significantly increased, a significant rise in Cytc; Caspase-3and Cytc mRNA expression quantity extremely significant increase. HE compared group and HF, Caspase-3significantly reduce, Cytc significantly reduced, Caspase-3and Cytc mRNA expression was significantly decreased. Conclusion: 1. Long term high fat diet induced obese rats, lipid metabolic abnormalities, can cause testicular tissue structure of rats changed, total sperm count reduced, activity rate, deformity rate, but also can make sperm apoptosis increased significantly.2. Long term high fat diet induced FSH, LH, T level in serum of rats decreased, and lipid peroxidation enhanced, probably because the increase in free radicals in the body.3. Long term high fat diet can induce apoptosis of rat spermatozoa increased, Caspase-3, increased Cytc expression in testicular tissue, aerobic exercise can reduce sperm apoptosis in rats, Caspase-3and Cytc expression decreased, suggesting that aerobic exercise through the Cytc, Caspase-3to participate in the intervention of mitochondrial apoptosis pathway in obese rats in vivo sperm apoptosis.4. Sperm apoptosis in rats induced by high fat diet increased, but the high fat fed simultaneously applied after swimming, rat sperm apoptosis rate decreased, suggesting that swimming can effectively alleviate the apoptosis of sperm in rats.
