Objective: To choose a better effect of two different way of leucine supplement in old rats skeletal muscle protein metabolism and to make suggestions of health problems in old people.Methods: Male old rats(n=14) undertaked overload induction and leucine supplement intevention. Then randomly divided into three groups.Control group(n=5),Leucine 1 group(n=5),Leucine 2 group(n=4).Leucine supplement(Leu1 group 0.2ml/kg BW;Leu2 group 0.4ml/kg BW,control group supply saline instead.) After 1week of intervention, rats of all groups were sacrificed.Insulin level was measured from plasma. P-m TOR/m TOR, P-p70s6k/p70s6 k and P-rps6/rps6 expression were measured by Western-blotting from skeletal muscle.Results: After 1w, compared with control group,plantaris and solues mass in Leu1 and Leu2 group significantly increased(P <0.05) and plasma insulin level significantly inreased in in Leu1 and Leu2 group.And compared with left side skeletal muscle,P-m TOR/m TOR, P-p70s6k/p70s6 k and P-rps6/rps6 expression were all significantly increased in right side of Leu1 and Leu2 group.(P<0.05).Compared with Leu2 group, P-m TOR/m TOR, P-p70s6k/p70s6 k and P-rps6/rps6 expression increase all have significantly difference in Leu1 group.Conclusion:Overload induction surgery causes increase of skeletal muscle mass.And leucine supplement in old rats has positive effect in promoting protein synthesis of skeletal muscle through activating m TOR signaling pathway. |
