Objective: By establishing the middle-aged and old rats 8 weeks aerobicexercise and curcumin intervention model, constant load test movement and curcumin rats soleus AMPK activity changes after intervention and autophagy related gene expression, and using HE staining to observe the histopathologic changes of skeletal muscle of rats, to explore movement combination of curcumin induced AMPK activity changes on the influence of the middle-aged and old rats skeletal muscle cell autophagy. Speculated that the movement of autophagy and the mechanism of the curcumin prevent Sarcopenia.Methods: 40 healthy male with batch 21 months SD rats, SPF(Specific pathogen Free, SPF) level of animals, cage. Adaptive feed for three days, randomly divided into 4 groups, were randomly divided into blank control group(BC, n = 10), quiet curcumin group(QC, n = 10), a constant load exercise + curcumin groups(EC, n = 10), a constant load movement(NE, n = 10). Each component 2 cage feeding, each cage 5 only. Movement group rats using treadmill exercise, establish curcumin animal experiment model combined with aerobic exercise training intervention mode, the slope is 0, 15 m/min speed x 60 min(load intensity is equivalent to 70% VO2max), curcumin dose of 100 mg/kg weight, continuous training for five days a week, a total of 8 weeks training. 20 hours after the end of the training time remove the hind limbs of rats soleus, calculate the relative weight of the rats soleus(SI), using Elisa method to detect the soleus AMPK activity change, by real-time fluorescent quantitative PCR detection AGT7 mRNA, LC3- b mRNA, LAMP- 2 mRNA expression, and from the aspects of morphology using HE staining to observe the histopathologic changes of rat skeletal muscle.7)Results:1) Each weight of rats were significantly lower(p < 0.01); After the experiment, QC group, NE and EC weight of rats were significantly higher than in BC group(p < 0.01), NE group is higher than the QC group and the EC group(p < 0.05).8)2) Compared with the BC group, QC group, the NE and EC group SI values are higher, and it has very significant difference(P < 0.01); NE group is higher than the EC group, with significant difference(P < 0.05).9)3) Groups of middle-aged and old rats soleus appeared different degree of histopathological changes associated with aging, relative to the BC group and QC group, the NE and EC group woven abnormalities like appears the frequency is low, among which the best NE.10)4) Compared with the BC group, QC group, the NE and EC group AMPK activity increased significantly(P < 0.01); Soleus AMPK activity is highest among them, the EC group, compared with NE, QC group, has a significant difference(P < 0.05).Conclusion:1) 8 weeks of constant load aerobics, curcumin and combining both intervention can maintain skeletal muscle weight to a certain extent, improve the pathological changes of skeletal muscle tissue, its mechanism may be through the activation of skeletal muscle AMPK, improve the middle-aged and old rats skeletal muscle AGT7, LC3-b, the LAMP-2 gene expression.2) Curcumin and constant load aerobic exercise may be induced by activation of skeletal muscle AMPK autophagy this mechanism to maintain skeletal muscle mass and improve skeletal muscle lesions.3) Aerobic exercise than curcumin and eight weeks of constant load of curcumin in combination with a constant load aerobics to ease the loss of skeletal muscle mass and improve the role of the muscular tissue pathological changes, aerobic exercise can make the skeletal muscle may be due to the constant load maintains a more adapt to the level of autophagy.
