Objective: By detecting the rat soleus muscle IGF-1, change Pi3 k / Akt / m TOR protein expression of its content to cell apoptosis, explore curcumin, constant load exercise,incremental Exercise on Pi3 k / Akt / m TOR signaling pathway.And comparing curcumin, constant load exercise, incremental load exercise, combined with constant load exercise curcumin,curcumin combined rat skeletal muscle aging effect of exercise on increasing load delay.Methods:The same batch of 18 healthy male SD rats were 60 months of age(555.61 ± 57.44g), SPF(Specific pathogen Free, SPF)level of animals, housed separately. Adaptive feeding for three days, were randomly divided into six groups, were randomly divided into control group(Control group, BC, n = 10), curcumin(Curcumin group, QC, n = 10), constant load exercise group(Constant load exercise group, CE, n = 10), increasing load exercise group(incremental Exercise group, IE, n = 10), a constant load exercise + curcumin group(constant load exercise curcumin group, CC, n = 10), incremental Exercise + curcumin group(Incremental Exercise + curcumin group, IC, n = 10). 2points each to cages, five per cage. Exercise group rats using treadmill exercise, the establishment of a constant load and< strength 19 m / min × 60 min, increasing load exercise intensity Referring to Table 1-2. Slope of 0(equivalent to Exercise Intensity 70% VO2max), five days a week for training,two days of rest, training a total of 8 weeks. After the end of the last 20 hours of training to take blood, soleus muscle was measured IGF-1 content using Elissa method, using Western Blotting Detection Akt, Pi3 k, m TOR protein expression, and the situation of skeletal muscle cell apoptosis by TUNEL assay.Results:(1) Muscle apoptosisin rat soleus smallest index is constant load exercise group, followed by the curcumin group,apoptotic index is the largest group increasing load. Compared with curcumin group and constant load group, there was a significant difference P < 0.01; compared with curcumin increasing load was significant difference P < 0.05.(2)compared with the group and curcumin group constant load,increasing load in serum IGF-1 content was significant difference(P<0.05).(3) Compared with the control group and curcumin group, there was a significant difference(P<0.05)muscle content of IGF-1 in rat soleus curcumin constant load group. There was a significant difference(P<0.05) between the group and the constant incremental load exercise intensity group. Expression of content(4) of the rat soleus muscle Pi3 k between curcumin and constant load exercise, there is a very significant difference P < 0.01. There was a significant difference between a constant load constant load group and curcumin group. P<0.05.(5) Compared with the control group,curcumin, curcumin group set constant load, constant load exercise group, curcumin increasing load exercise group, rat skeletal muscle increasing load exercise group Akt content were significant differences in P<0.05. Compared with curcumin,increasing load group there was a significant difference P<0.05.(6) Compare m TOR rat skeletal muscle, compared with the control group, curcumin Incremental Exercise has significant difference,P<0.05, curcumin has a very significant difference P<0.01; compared with curcumin group,increasing load and constant load group group were significantly different,P<0.05.Conclusions:(1) Constant load of aerobic exercise can promote the IGF-1, the expression content Pi3 k, Akt, m TOR increases in rat skeletal muscle, delaying apoptosis rate.(2) Incremental Exercise makes rat skeletal muscle IGF-1, the expression content Pi3 k, Akt and m TOR is reduced, accelerate the rate of apoptosis.(3) Curcumin can effectively improve the body’s antioxidant activity, change IGF-1, the expression levels of Pi3 k, Akt, m TOR, slowing the rate of apoptosis, but also may be associated with cell regeneration related.
