By analyzing the economic density differences, regional economic difference and the evolution of the spatial pattern of regional economic were studied. Based on analyzing its impact on labor productivity, the impact of the economic activity or element was assessed, which provided a new direction for the study of regional economic differences and promoting regional coordinated development. Therefore, 279 prefecture-level cities were taken as research unit, the years during 2004-2013 as study period, and ArcGIS, Stata software as analysis platform, from the perspective of economic density, the effect of economic density distribution on labor productivity was studied systematacially in this paper by using geo-spatial analysis and spatial econometric model.The results show that:(1) The characteristics of spatial differentiation and agglomeration of economic density were obvious. The spatial differentiation characteristics of the economic density among different prefecture level cities in China are obvious. The differences existed mainly in the direction of east and west, north and south. Besides, the trend of economic elements to local spatial concentration was obvious. Based on the analysis of spatial distribution pattern, the spatial agglomeration model of economic density was identified furtherly by determining the spatial relationships among prefecture level cities, which was the coexistence of spatial agglomeration and diffusion of economic density. The core areas were mainly distributed in the Yangtze River Delta, the west side of the Taiwan Strait, the Pearl River Delta. During the study period, there was a certain space contraction and showing a trend of intensity. At the national scale, the spatial diffusion of the high value of economic density from the eastern city to the central and western city was obvious. The labor productivity also had the characteristics of unbalanced spatial distribution, obvious polarization, and stronger spatial dependence.(2) After considering the endogeneity and spatial spillover, economic density had a significant effect on labor productivity. Under the premise of not resolving the problems of endogeneity and not considering spatial spillover, it can be found that the economic density distribution had a positive effect on labor productivity by the simple regression analysis, but the effect was not significant, linear regression linear regression effect was poor. Based on spatial panel regression analysis, it showed that the density of the third industry had a significant positive effect on labor productivity, and the regression coefficient was more than 0.34, but the regression coefficient was too large compared with the previous studies. However, compared with the previous studies, the regression coefficient is too large. It can be found the existence of economic agglomeration effects by introducing the urban built-up area of instrumental variables and considering the spatial spillover effects of the labor productivity among prefecture level cities, especially secondary industry-density, tertiary industry density, and spatial spillover effect had a significant positive effect on laber productivity.(3) The influence of economic density on labor productivity has the spatial heterogeneity. In different area, the influence of economic density on labor productivity was not homogeneous.There were agglomeration effects, but obvious spatial differences the four regions in China. In the eastern region, the concentration of the output element was not economic, but the tertiary industry density and the spatial spillover effect of labor productivity among prefecture level cities are more and more conducive to the improvement of the level of labor productivity. In the central, western and northeast region, effect on density of investment and per capita fixed asset investment on labor productivity were more obvious. On the urban agglomeration, the third industry density and employment density had an obvious effect on laber productivity in urban agglomeration of the higher development level, and the output density had a great impact on labor productivity in urban agglomeration of the lower development level.
