Since the financial crisis in 2008, the world economy is pessimistic and rising unemployment. Many countries have introduced various policies which to encourage technology entrepreneurship, regional economic growth and full employment. The birth and growth of high-tech enterprises can effectively promote regional economic sustainable growth by the allocation of resources, technological innovation, system innovation, industrial agglomeration. How to improve the level of regional science and technology entrepreneurs to achieve rapid industrialization of scientific and technological achievements is increasingly becoming the focus of the business community and theorists. In this context, the mechanisms of regional science and technology based entrepreneurial ecosystem and the policy is important realistic and theoretical research.In response to these problems, on the perspective of complexity science to explain the mechanism of regional technology start-ups based bottom-up logic analysis. The regional science and technology based entrepreneurial ecosystem is a complex system which consisting of Multi-Agent. Based on complex system theory and technology entrepreneurship theory, a Multi-Agent model of regional science and technology based entrepreneurial ecosystem is conducted which include the government, the incumbent companies, intermediaries, financial institutions, universities and research institutions. The model highlights the role of interaction between heterogeneous agents in the evolutionary process of regional entrepreneurial ecosystem. Based on technological entrepreneurship in Nanjing, Netlogo simulation modeling is carried out to analyze the effect of parameter variation on the numbers of technology firm in the region. The results indicate that (1) Entrepreneurship policy system of government is more conducive to fostering regional technological entrepreneurship system than a single policy measures. (2) When the government to introduce entrepreneurial talents, relevant intermediary organizations and venture capital institutions should be introduced. (3) Government venture capital funds are more effectively promote to improve the levels of regional scientific and technological entrepreneurship by PE/VC institutions investment for technology entrepreneurs. (4) Good entrepreneurship culture and regional industry-university-research cooperation network are essential to successfully build an efficient entrepreneurial ecosystem.In the case study, the simulation studies’ conclusions were verified which taking Nanjing as an example. Based on these studies, the regional science and technology based entrepreneurial ecosystem in Nanjing was studied which included the situation of the technology entrepreneurship, the construction of the regional science and technology based entrepreneurial ecosystem, the introduce of the advanced experience and relevant policy recommendations.
