The Alpinia coriacea is a kind of plant in the genus Alpinia(Zingiberaceae). It is an endemic plant only distributed in Hainan Province, is usually found in open forests. Fruits and roots of the Alpinia have lots of medicinal values. Generally, it has the effect of aromatic stomach, warm in cold, anti-inflammatory analgesic. A. oxyphylla Miq, A.officinarum Hance, A. galanga willd were written in Chinese pharmacopoeia. There is no any report about chemical constituents and pharmacological activities of A. coriacea. In our preliminary study, we discovered that the ethanol extract of A. coriacea showed significant pharmacological activities. In order to rational development and utilization of A.coriacea, we research on the chemical composition and pharmacological activities of A.coriacea for the first time. The study provides theoretical basis for its further development and utilization.30 compounds were isolated from A. coriacea by means of silica gel column chromatography, Sephadex LH-20, ODS, HPLC etc. which. The structures of compounds were identified by spectrum technology, such as IR, UV, NMR spectrum, and so on. The compounds were identified as: alpicoriacine A(1), zedoalactone B(2), zedoalactone A(3),zedoarolide A(4), 7-hydroxypiperitone(5),(E)-labda-8(17),12,14-trien-15(16)-olide(6),pinocembrin(7), apigenim(8), izalpinin(9), kaempferol(10), 5, 7, 3?, 4?-tetrahydroxy-flavanone(11), 3, 5-dihydroxy-7, 4'-dimethoxyflavone(12), quercetin(13),5,7,3 ?,4 ?,5 ?-pentamethoxyflavone(14), 4-hydroxybenzaldehyde(15), anisic acid(16),cinnamic acid(17), syringate(18), methyl syringate(19), 3,4-dihydroxybenzoic acid(20),?-tocospiros A(21), stigmast-4-ene-6 ?-ol-3-one(22), stigmast-4-en-3-one(23),7-keto-?-sitosterol(24),(22E)-5?, 8?-epidioxyergosta-6,22-dien-3?-ol(25), stigmasterol(26), stigmast-4-ene-6?-ol-3-one(27), stigmast-4-en-3,6-dione(28), ?-sitosterol(29), and?-daucosterin(30). Compound 1 was a new compound. Compounds 1-6 were terpenoids;Compounds 7-14 were flavonoids; Compounds 15-20 were phenolic acids; Compounds22-30 were sterides. Compounds 1-5, 8, 12, 14, 16, 18-19, 21-25,and 28 were isolated from the genus Alpinia for the first time. All of the compounds were isolated from A. coriacea for the first time.Compounds 2, 6, 8, 15, and 26 exhibited activity against M. tetragenus with MIC values of 10 ?g/mL, Compounds 2, 4, and 25 exhibited activity against E.coli with MIC values of 5 ?g/mL. Compound25 exhibited activity against S.albus and B.subtilis with MIC values of 10 ?g/mL.Compounds 2, 6, and 27 exhibited weak cytotoxic activity against Hela ?SMMC-7721 and A-549. Compound 23 exhibited weak cytotoxic activity against Hela.The essential oil of A. Coriacea exhibited weak cytotoxic activity against Hela and A-549.
