Research On The Vivo-antioxidant And Vitro-antioxidant Effect Of Kudzu Flavonoids In High-Fet Diet-Fed Mice

Objective: Studies have shown that long-term high fat diet can lead to a large increase in fat and promoted fat oxidation, generated excess free radicals and damage the organism and.disease. Many studies have found that flavonoids has the very good antioxidant effect, but little report about flavonoids reduce oxidative stress injury induced by high fat. Therefore, this paper mainly studied the effect of Kudzu Flavonoids on antioxidant index of high fat diet mice and Scavenging activity in vitro,Evaluation of radix puerariae flavonoids antioxidant capacity.Methods: 50 female ICR mice were randomly divided into 5 groups, normal group,high fat diet group,high fat+ 0.2% Kudzu Flavonoids, high fat+ 0.4% Kudzu Flavonoids,high fat+ 0.8% Kudzu Flavonoids, feed eight weeks, Weekly weighted body weight one times. Dissect mice and Culling heart blood and separate serum, Organs and intraperitineal adipose tissues were weighted at the end of experiment. Analysis related activity of antioxidant enzymes of serum and liver(T-AOC,SOD,CAT,GSH-Px,MDA).The determination of scavenging free radicals scavenging ability in vitro. Evaluation of radix puerariae flavonoids antioxidant effect.Results:1)Compared with the high fat diet group, the body weight and the IPAT weight of Kudzu Flavonoids groups were significantly reduced.2) Compared with the high fat diet group, Kudzu Flavonoids significant improved activity of antioxidant enzymes of T-AOC,SOD, CAT and GSH-Px of the mice liver and serum and significantly lower in liver and serum lipid peroxidation product lipid peroxide(MDA) content.3) Kudzu Flavonoids on DPPH free radicals · OH,2O·has the very good scavenging effect in vitro and has positive dose-effect relationship.Conclusion: Kudzu Flavonoids effectively scavenging free radicals, Can improve the body of a high-fat diet causes oxygen stress, regulate the body's oxygen balance.
