In the research,using the waste of cornus wateri as the material.Selecting the method of extra high voltage,ultrasonic wave and microwave to extracting cornus walteri pigment,through comparison of the three methods,we can obtain the preferred method for extracting cornus walteri pigment,and then purifying the crude pigment,studing the stability and analyzing the component.Using the ethanol as extraction liqid,measuring absorbance of the walteri pigment at the wavelength of 380~620 nm to get the absorption spectrum of cornus wateri pigment,and determining the maximum absorption wavelength is 540 nm.Through single factor and orthogonal experiment to geting the optimum conditions of the method of extra high voltage:the ethanol concentration is 50 %,liquid ratio is 1:40,time is 20 min,extraction pressure is 200 Mpa.The optimum conditions of the method of ultrasonic wave: the ethanol concentration is 50 %,liquid ratio is 1:40,extraction time is30 min,ultrasonic power is 120 W.The optimum conditions of the method of microwave: the ethanol concentration is 70 %,liquid ratio is 1:40,extraction time is 20 s,microwave power is 350 W.Through the comparison of three different extraction,including the advantages of extraction methods,the aspects of material,the aspects of extraction effect and the aspects of extraction time.Founding that comparing with the other two methods,the method of extra high voltage has its own unique advantages,and it is a preferred method of extracting cornus wateri pigment.On the aspect of purification and analysis of the component,through experiments of static adsorption and desorption,choosing NKA-9 as the best choice resin of purifing cornus wateri pigment.Through single factor and orthogonal experiments to get the optimum conditions of dynamic adsorption: the pH of the sample solution is 6,the absorbance of the sample solution is 0.7,adsorption time is 20 h,adsorption temperature is 30 ?.At the same time,studying the two important factors that influence the effect of the elution,including the concentration of ethanol and flow rate of desorption,experiments show that the best choice is that the concentration of ethanol is 70 % and the flow rate of desorption is 0.5 ml/min.Through the comparison of the spectral characteristics and color value,we can confirm that the process of purification has retained the original composition of good pigment,and at the same time,the color value has increased significantly.Through the study of the stability of the cornus wateri pigment,we can confirm that cornus wateri pigment has thermal stability and light stability,pH affect its stability seriously,common food additives and metal ions make little affect on it.Using the technology of HPLC-MS to analyze the component of cornus wateri pigment at the first time,we can confirm that the main component of cornus wateri pigment is quercetin possibly,and cornus wateri pigment may also contain other pigments,like anthocyanin.
